Damn, I was so careful when I inoculated my spawn jars though. I cleaned my whole apartment, sprayed lysol everywhere, showered, facemask, gloves... the works. I wonder where it went wrong.

I hate trimming the liner I've made a few recent tubs without a liner... mad doesn't use a liner he's got the voodoo where he gets no side pins haha... knocking up with a syringe without a doubt man

Syringes aren't clean, use agar in a SAB, practice your procedures, don't reach over your work ect.
If problems still go over your filters, up your PC time keep troubleshooting eliminating one vector at a time.

always have some uninoculated control jars to make sure your filters are good. Up the PC times to at least 2 hours, and get a SAB and do agar. I can dig around and show you what spore syringes look like if you want lol!

So true tho. I knew something was up. I shouldn't have second guessed myself haha. They didn't have excessive fuzzy feet and the caps looked big enough for their actual size. Fuckin skinny trich pins lol

It sounds like you meant well with the precautions, but did you flame sterilize the needle before each injection? Also did you swab the injection ports with isopropyl immediately before? Those are a couple things I've always done. It is riskier with MS and grains, but it is still possible.

Yea, I flame sterilized, but I did not swab with isopropyl. I didn't even know that was necessary.
The trich pins is also news to me. I read a few threads on thin pins and none of them mentioned anything about trich causing them.

No wonder why agar is such a big part of growing. I always thought it was because it saves money on syringes lol. Had no idea it was more sterile.

Agar forces the culture to grow in 2 dimensions where you can identify and transfer away from contams, ensuring you have a clean inoculant. Several other benefits like cloning and growing countless grows from a single drop, or using a swab or loop to grow from your own prints, storing live cultures and proven genetics for years and so on good stuff

Yeah, working with agar is a sure fire way to get clean inoculate. Especially if you do repeated transfers of high performing strains. You should look into isolating. That way you can get some "cream of the crop" fruits. And, it was all under your control. I wish I would have done that when I first started. I started with agar immediately, but I wish I would have know about isolating, different agar recipes, do's and don'ts. Instead, I let my agar grow and inoculated via "tiger drop" (something else for you to know).

Cloning is cool but isolating is tedious as fuck. The time and effort invested is not worth it unless you are doing it as a side project and don't care about how long it takes. Get 20 clones and keep the best one. Have some fruits in the meantime.

....that's what I meant lol. Ot is good information to know though. I figured you as an isolating guy Pasty. So you just took a bunch of clones, expanded them while keeping a master, and did a test run with all your other clones in order to find the best one, and then when you found it, you went back to the master to create more?
Also, maybe you can/will answer this for me Pasty...why isn't senescence an issue when it comes to working with agar and isolating like it is when you do a G2G. I've read some accounts of people using upwards of 20-30 plates before they got their isolate perfect.

It is an issue. I am careful to keep my master cultures from growing out too far between slants. Obviously a true isolate will live longer than a clone but a clone can still last a long fucking time.
I do have a true isolate. It's a PE that produces terrible yields but can drop spores. But my best clones are still worthy producers, my RW F2 clone will dump out 10+ dry oz per 66 quart tub on the first flush.
