Ok im getting the hang of it....had a couple of failed transfers that went moldy/bacterial but i now did two clean transfers.
What still confuses me is...my Myc is really fluffy. At first i thought its some kind of mold but it stays white and only grows kinda slow. Do i just need to make more transfers and wait until i get a less fluffy Myc that grows faster?

Thanks so much for this tek Pasty! I have been researching and wanted to get into agar. I just finished PCing my plates and waiting overnight for them to cool so I can inoculate them with APE spores. These plates will be used to start some WBS jars for my first bulk grow.
It was so freaking simple and crossing my fingers that they successfully colonize. I just created my Still Air Box and have a HEPA filter/UV running in the room overnight to prepare for adding spores. So excited!

So I'm trying to make a really big batch at once, roughly 4.7x original recipe size. My shit is super milky and hardening very quickly. Like complete solid gel after sitting at room temp for like 2-3 minutes.
Do the potato flakes not scale up along with the rest of the recipe? Tried a new color of agar because my others were out and fear I may have gotten one with sugar added or something.

Would a glove box help against high spore count?
I used a SAB as an experiment to check contam vectors by pouring test agar petris, and almost all plates fell with black mold. I'm convinced it is high spore load as I live in a tropical country, high humidity, rainy season. I was thinking a glove box would help. My reasoning is that it would reduce my contam rate if I bleach bomb inside (kill the air borne spores at least) and then quickly pour the agar plates.

YES of course it will help!!
the key of a glove box\SAB is the still air so u can bleach to kill contam but the most important thing to do is wet the walls of your SAB
With wet walls the contam will be stuck in the water against the wall and they don't bother you anymore (usually)
and yes also the fast u do it the better

The problem with glove boxes is they push air around the inside because of the gloves. Airtight or not.
A SAB should be sufficient. What kind of room are you using your SAB in? How long do you let the air settle after killing the ac and fans?
I'm not saying a glove box won't work, but it shouldn't really do anything a SAB can do when used properly. The spore load will still be there. Maybe purchase a small air purifier to run while your not doing clean work. Maybe try to reduce the problem, then just working around it.

still air is key. if the air in your SAB is still and all mold spores stuck to the wet walls it doesnt matter how dirty your room, air or feet is. You need to move smoothly so you dont stir up the air, plan your movements before doing them. and just practice.

But if in a glovebox, as opposed to a SAB, then I can at least reduce the spore count with bleach since the spores will be, for the most part, contained. Problem is that I worked on the SAB and planned my movements, and the air is still, nevertheless got the contam, which leads me to find another viable way to do this. I'm talking about pouring which is like 3-4 second exposure altogether, oh, and i'm in a pretty much, what you could call, a jungle type environment.

Anyone can get good at the SAB. If you can't do it in there the glovebox won't be much easier. You still shouldn't put an iso-soaked glove over your work no matter how you're doing it.

Noc, you're an expert on agar. What say you? Do I need slides or something to check or do you know what's up?
They solidified enough in like 20m to do a damn tiger drop.

Anyone have any UK replacements for mini rounds? Doing a test run with some pp5 that are clip down lids instead and hoping they work.
My last batch grew nothing in a week, not even contams :o

You should try www.ampulla.co.uk for see-through pp5s in europe. They also have a wide variety of glass jars to choose from. They are the cheapest source for glass jars I could find, by far. I think their quart jars are still out of stock though, unfortunately.
