Fischer's Zombie Cube Grow Log

Hello everyone.

After being dissapointed by shitty genetics on my last grow that I started on 6/20, ive decided to start fresh.

Today i am going to be making several plates and inoculating with spores and dried tissue. (Spores are a safety net, the real experiment is to see if i can have success cloning fruits that are about 2 years old)

Im going to take several approaches while doing this to see what works best.

My plan is going to work as follows.

Im going to make 20 dishes.

-5 plates will be from peroxidated dried material, using only stipe

-5 will be from the stipe of mushrooms that im going to rehydrate in sterile water, and then once rehydrated i will clone by taking tissue from the torn stipe

-5 shrooms will be rehydrated in sterile water, peroxidated, then the sample will be taken from the stipe

-2 will be from a non-peroxidated, non hydrated stipe

-3 will be spores from a print.


The strain of both the spores and dried material is Mazatapec, my favorite strain by far.



I am going to begin preparing my flowhood, autoclave, and agar (im using PDA) now. Will be posting progress as it happens.

I'll put my chips on the non-peroxidated rehydrated stipe for ease of isolation, but I'm tied between whether those or the spores will provide the faster result. How old is the spore print, out of curiosity?

I don't think bleach slows mycelium down the way H2O2 does, but the contaminants will still take a big hit. What results will you be looking to study from this experiment?

If you're looking to get clean growth on your agar, have you considered trying the agar sandwich method? Or perhaps pouring hot agar on top of the growth and letting the mycelium grow upwards through it? It would cut down on your number of transfers until clean growth, which sounds beneficial when working with dehydrated tissue. I've heard contamination can be a big problem when dealing with it.

In terms of results, im trying to find a viable, reliable way of cloning tissue from a dried specimen.

Obviously it would have different applications, in this case these are fruits from a great grow i had that was an aggressive colonizer and one that I wished i had cloned at the time.

It seems like bacteria is a big problem when trying to clone dried fruits so maybe after seeing how growth(if any) goes, i will use the sandwich method.

Ive already used a low nutrient PDA recipe so hopefully it works to my advantage. Time will tell.


In the meantime the stipes are soaking and im waiting for the agar to cool.

Well i inoculated fully and was wrappimg the spore inoculated plates with parafilm and knocked all of thr spore plates over.

So id say now they have just as good of a chamce as the dried tissue :blush:
