Making LI from Petri dishes - Problems.

I have been making LI in a mason jar/blender for a year now (using teks found on this forum). I am noticing that the petri dishes I use that have been in the fridge are weaker than ones that haven't had their growth slowed.

What can I do to make the mycelia the most robust in this situation? I have been bringing the dish out to room temp 12-24 hours before I cut it up and use the entire dish in 60mm of water. About 15mm used in each 4 quart myco bag via syringe.

If you like to use LC i recommend to use GLC in the future for long term storage. You can take some colonized agar and transfer it to a small grain jar. Inject sterile distilled water when the jar is completely colonized and suck it into a fresh syringe. You can store it in the fridge for later use.

Taking it out early does nothing to re invigorate the mycelium. Use fresh mycelium for LI. The older the culture the more glycogen depleted the cells are and hence lag.

GLC is garbage. Grains are not completely sterilized even at 2 hours at 15 psi. We sterilize for a window to colonize the grains. GLC is asking for trouble, maybe not often. But it's just plain shit given the alternatives

Try Munchausens Blenderless LI tek in conjunction with pasty plates. 

They work amazingly well together!!!

May be more risk but i never had a contam with GLC (150+ GLCs).
You may call it garbage but if you use a small amount of grain chances for a contam are really low IME. Whats your favorite method for long term storage TC?

OK. So I'll need to plan out fresh petri dishes. Thats a bummer.

What if I put petri dishes in the fridge when they are barely started, then take them out a week before to grow them out, is that reasonable?

I'm more interested in doing agar work infrequently, because I clean my whole place before I use my still air box. So thats why I'm brainstorming a process that can allow less tedious sterile work.

You don't need to clean your entire house man.  If anything this will kick up particles into the air, which is worse. 

Just use a room where the air is relatively still, and work only in the SAB.  Use sterile gloves if possible and normal sterile procedures (flaming tools etc).  That's it really.

I guess I don't find it all that tedious or cumbersome. 

Have you checked out Pastyplate tek?  It really makes things MUCH easier, especially if you don't like pouring agar in a SAB.  You can make tons of plates and just PC them in the container they colonize in.

If you want good results, you need to keep your cultures young and vivacious. Everytime I make a batch of LI for my grains, I do 5-10 pasty plates. it takes 15 minutes in the sab to do all the transfers. If you don't have time for that, there is always the pf-tek.

I make batches of 4 pre-sealed large myco bags pressure cooked. I love noccing them up with a syringe. I like LI for safety. So here I am. Works like a champ with fresh petri dishes. I'm just trying to make a good system. I'm liking my idea of cooling a just started petri.

I've been doing about 30 of those a day for a long time and have tried many different methods but the most successful for me has been colonizing half gallon jars from agar wedges, then G2Ging each of those into 10 bags. Colonized grain jars can chill in the fridge for atleast a few months before they get expanded. I do all of my work infront of a large flowhood though so I understand if liquid is your only option. Just my 2cents!
