Hi,

Was going to attempt a second transfer to a new plate with this bit of tampanesis mycelium.
I don't have any experience with this strain and I can't see any stones on the plate, but would anyone know the best place to take a cutting for the next plate? Say the 12 o'clock position, 3, 6, or 9?

You could wait until you can see stones growing on agar and transfer the stone forming sectors to another dish.

Okay - I guess they would look a little darker colored?
It is something that I can't seem to find much information.
I checked out StoneSuns comprehensive guide, but it looks like he has just flat out grown plain looking mycelium and got good results with that alone -
In which case I would just transplant the fastest growing bit?

I'd transfer now. 12 ,6, or 9 would be fine. Stay in the main circle. That secondary spot at 3 looks different. Although that's probably ok too.

If you have enough agar plates you definitely could transfer now to separate some genetics from each other. If you want to save time you can transfer brown spots or as i already wrote wait until you see sclerotia forming and transfer them.

Its like isolating or cloning Cubensis. Cloning can save some time and give you good results. If it is a long time project you can try to isolate a nice strain. It would take some time and petri dishes of course..
