Spotting Hidden Contaminants on Agar

been doing lots of agar work lately and wanted to share some tips that have been useful for me, and invite others to share any techniques they have for identifying hidden contams in agar cultures

Of course, if you have a microscope handy, you can always physically inspect the myc. you can also do much of the same with a large magnifying glass. i have a magnifying glass with LED ring just for this purpose.

another trick is if you want to take macro shots with any camera, hold the magnifying glass up to the plate and then allow your camera to focus (ideally use manual focus) on the plate through the glass. If you have a decent camera, you can use this technique for lots of stuff. I like to take a big blown-up backlit shot using a lightbox and magnifying glass, then adjust the exposure and other settings in Photoshop/Camera Raw to make the features stand out the most, then i use a high resolution screen (2-in-1, tablet, laptop, etc) to display the image above my workspace. this is extremely helpful when trying to make VERY small transfers, make precision transfers of hard-to-spot characteristics, as i can look to the screen as i take the sample

but the tip i use most of all is to bounce light off of the surface of a plate, holding it at an angle that reveals features (often bacterial contams) which are not visible to the naked eye head-on. this is easiest to notice on uncolonized parts of the agar, which is useful for choosing which section of the leading edge to transfer. but by reflecting light off of the surface of the dish, it is also possible to see splotches and blemishes on the colonized sections of the dish as well. Often, it could just be water that has condensed on the surface, but this is often a vector of contamination, so its probably a good idea to avoid transferring questionable sections

i will post if i think of any more, and i look forward to seeing any similar techniques others use to spot hidden contamination in their cultures

wow thats some funky lookin growth, any background on it? how many transfers in, source, etc?

i was going through some old plates a few days ago throwing out all the unneeded stuff and came across a really crazy plate. it was just normal rhizo growth before putting it in storage, then a few weeks later i noticed this crazy crackle pattern growth
