Smelly peroxide petri's
    #2360375 - 02/20/04 06:42 PM (12 years, 9 months ago)

We innoculated a series of 10% peroxidated mda petri's using an old full grown azure mycelia plate, a week later there is no visibe growth of either mycelium or bad molds. Nasty bacteria smell is present despite the high peroxide concentration.
Is there any way to save clean myc from these cultures?
Thanks in advance,
-phloid

something has gone wrong, of course. a peroxide level that high is not advisable. and if it is that high there is no way in hell that bacteria got on there and lived. i say try again but do not use more than 10ml per leter of agar, at max. that's closer to 1/100, not 1/10. even that is a lot by most standards.

According to R. Rush Wayne, Ph.D. anywhere from 0.018% to 0.036% peroxide in agar can be used. This is the same range as adding 6-12 ml of 3% peroxide per liter batch. Of course, the higher the peroxide, the slower the fungi will grow.

I agree, if in fact the agar had 10% peroxide that is way too high for what he needs it for.

H ow do you explain bacteria survived? What about transfering to wood chips>? Maybe pasteurized?

My friend prepared some agar for her first time and had a couple of problems. She used 150ml potato broth 350ml distilled H2O, 9g AGAR, and 10ml honey. She pc'd this for 17 min. @ 15 psi. After the agar broth had cooled to about 100oF she added 50ml 3% H2O2. Before allowing the solution to solidify, it was poured into 12 different 4oz ball jelly jars which were sterilized (3$ at WALhell for a case of 12!!!). This of course made the Agar much to thick, but since it was the first time she tried AGAR, my friend went ahead and placed cleansed fruit bodies on the dishes. All did well, except they seemed to only colonize 1/10th of the agar, in terms of depth. The whole top however was completely colonized in under a week. Don't know if this helps any, but it was my friends experience.
