The other Plates that i innoced with the Spore Sollution starting to look kinda bacterial too...guess i wont try using a Spore Vial for agar again.

Trusted cultivator just recently posted a good method for streaking a syringe with an inoculation loop, which seems like a good method when using possibly contaminated spore solution.  I haven't figured out how to post a direct link to his post but I'm sure he can help you out.

You can steam agar but I would probably go 2 hours.

Instant mashed potatoes = potato flakes. Potatoes provide specific nutrients required for the mycelium to reproduce cells (i.e. grow).

Any tool you can flame sterilize works. Ideal tools get hot fast and cool down relatively quickly.

The entire point of agar is that if contaminates are present, you can take a clean transfer to a fresh plate to 'clean it up'. It does not allow contaminates to hide, so when u go to put it on grains u know the culture is clean and not harboring any unseen nasties.

Agar is the most important tool in the hobby but nothing to be scared. It's actually quite easy. Check my signature for videos on agar.

Here you go dude, this may help you with your bacterial problem:

36fuckin5 said:
If you want to streak a plate, flame your syringe and inoculation loop. In your sterile area, put one drop of solution on the loop to cool it and the syringe. Then one more drop to actually pick up spores. Streak the plate with that.

But we really want growth starting from the middle of the plate so we can see the sectors and so that contams will be confined to one spot. Streaking is more for bacteria.

take your spores to an inoculation loop. if you have a print then flame the loop dip in agar, pick spores off print and then swab the dish

if you have a syringe flame the loop and drop a few drops onto the loop to cool it down and then one more drop to load it with spores and then swab that

Try using an inoculation loop. Flame the syringe needle and the loop then squirt onto the loop to cool the needle and the loop. Squirt another drop to prime the loop. Then swab the dish.

Then you can see growth immediately, see contamination easier, grab healthy mycelium easier, and never worry about spore drops rolling all over a dish giving you germination everywhere at the same time.

how often do you wash your pp5 LIDS? i hate having to change the micropore tape every time. since they're getting PC'd before each batch, do they really need to be washed?

thats exactly what i'm asking though. can you please define "need to be washed"? for example, if they were used over a plate that ended up showing a couple bacterial contams, do they need to be washed in the dishwasher before being used for another plate? I would think not, since i was under the impression that the point of agar is that bacterial contams will only grow across the flat plane of the agar surface and thus isn't supposed to contam the lid. not sure how much water my understanding holds though, and i'm a noob. could you kindly be a bit more thorough? :thanx:

.... They need to washed if they're dirty.

Visibly.

If there's shit around the rim you think could plausibly wick inside.  Bacteria growing on agar doesn't shoot nutritional juice all over the walls and lid of the plate..

if you don't know when something needs to be washed, you're going to have a bad time.

I don't know if it's possible to over think this anymore than it has been.

ya know, I know this is gonna sound a bit sketchy, but so far I haven't noticed anything visibly from doing it this way:

I rinse the pp5s after plopping out the agar puck. Throw everything in a tub with a bit of dish soap and water, and I'll leave them like that a few minutes.

Then I rinse them and wipe them off as I'm cooking up the agar. Yeah, the micropore is probably gonna stay wet. I have two layers on every hole, and I use a slightly smaller hole, about 1/8". I hardly ever change the micropore unless it's apparent something got to it, like a piece of agar or something.

I pour all the plates as the agar is ready, and wait until I stop seeing the steam rise from the plates. Then I top them all and let them set. I'll then put on the folded towel and the foil, and begin to load them in my PC. PC ~45min @ 15psi. Let them set again over an hour after PC. And I'll store them in a tote.

I have plates I made 2 months ago that I've been saving to use for agar sandwiches. They still look normal, just no condensation at all.

My guess is, the micropore dries out with the towel sufficiently to continue being an effective enough filter? I dunno.

Is this the trick to preventing condensation? They set long enough to become solid enough to not slosh, but almost immediately after placing the lids on the tubs, the inside of the tub began to fog up.

Thanks in advance!  :awesomenod:

First off these plates are awesome and makes agar work
Extremely easy and fun but I keep having a issue with
condensation.

I've let cool overnight, brought pressure up slowly.
But I keep getting it! Doesn't seem to affect the
growth but makes it hard to see.

Any tips???

wait a few days before innoculating them. Store them in a clean tote/ ziplocs untill you need them and the condensation should dissipate.

I've stored plates for weeks and they are still good to go

also if you get pooling in the plate you can use that to collect all the condensation and dump that out  before inoculating in your sab

Any water that's still there after that isn't enough to be a problem.
