Preserving strains

So my friend bought 6 strains(2 more on the way) and used them all and have 2 quart jars of each. They all look great, he's using whole oats. Gonna start doing g2g but only have a handful of jars left.
So now he's at a crossroads. Should he just grow one strain at a time and keep the rest in fridge? Idealy he would like to keep a master culture, would this be the best way to keep a culture? Or should he isolate on agar first? How do he know he has taken the best possible substrain? Should he inoculate agar with a single grain? A colonized grain jar should have the same DNA if inoculated from multispore right?
I know I am always asking newbie questions but I'm slowly gaining more and more experience. One day I will be able to provide information for noobs rather than leeching it. But until then give me all you got! Muahahah

Are these spores you are talking about? I think you can refrigerate them if you aren't going to use them for more than a year. Though it is debatable whether that is necessary.
I think the general consensus is that you should start on agar.
There is no need to inoculate agar using a grain. Inoculate it with your spores.
Once you have some fruits you can clone them.
If you have some clones worth keeping, the best way to preserve them is in a master slant.

Nah I'm not a huge fan of mycology LOL And that is very sound info and makes perfect sense to me. I was under the impression spores only lasted like 2 months. One of the syringes is cracked so I want to use those first, after I salvage those spores can I keep an agar culture for a year or so like a syringe? Even if it lasted a couple months, I could inoculate a new Petri dish to preserve it for an extended period of time? The end goal is to have my cultures stored so I can run one strain at a time. This is because I only have 20 jars and it would be easiest to g2g the same strain I think. Thanks again man! Oh and should I just order the cheapest agar agar I can find online?

If I were you I would start by reading Pastywhites teks. Then keep reading his all his other post. The reason I'm recommending this is because in a few hours reading not only would you find the answers to the questions you ask, but you would also find some in depth explanations that are wholly on point.
Fruiting from multi-spore syringes is fine if you're just growing out a few shrooms for you and your friends. If you're trying to store cultures of this strain and that one, plan on doing g2g transfers and whatever other high hopes/goals/dreams you may have, knowledge is going to be your best friend.

Tell your friend to watch RogerRabbit's "Let's Grow Mushrooms" videos ... Particularly the parts on cloning, agar and slants. The best way to preserve a strain is in a test tube slant kept inside a refrigerator.
At this point it would be best to fruit the mushrooms you have colonized and then put a few tissue samples from inside the stipe (stem) onto agar. Follow up by making a couple agar transfers to ensure the culture is clean, and then move a piece of the mycelium into a few test tube slants and store for long term usage.
Once you have a culture in a slant, as long as it is stored properly, it can be used for several years before needing to move it into a new test tube. This is how all professional mycologists and mushroom growers maintain their cultures.
Personally I wouldn't recommend going into isolation at this point in time since it sounds like you guys are just starting out. Cloning works really well and I'm sure you will be happy with the results you can achieve with a simple clone culture.
 For agar powder I would suggest using pre mixed from a mycology supply but that is really just a matter of personal preference. I like it for the simplicity of 'just add water'

Just as a follow up. Thanks everyone for the advice, I have spent countless hours researching and have decided to take the plunge. Although, for longevity, I found a way to store cultures in distilled water, their supposed to keep for up to 10 years?! What do you think of dh20 vs slants???
