good day lovely people,
I have a ton of jars going right now, and the last 100 or so cube jars ive done have all been a beautiful alpine white. Ive been working with several new species and lots of new races lately, and was fascinated by how different oyster, lions mane, shaggy mane, reishi, and active species myc looks, so cool
anyways, i nocced up 24 jars earlier this month to test new cultures (Limited MS) and noticed some unusual characteristics.
The PE6 culture was the single fastest ive ever seen, being on every single grain within 3 days, which was in itself suspicious. the myc is a grey color that is quite different from all my other jars, and the parts where the glass is touching the grains are bright yellow. The top is solid grey, where it doesnt touch glass. my first thought is bacteria, but ive never worked with this race so i wanted to get some additional opinions

Also, i have some pan. trop. (pan cyan?) jars going that seem to show two distinct kinds of myc, at least. never worked with the pan family before, so idk if this is normal, this is also limited MS

And this particular batch of AA+ seems to show some pink splotches, probably bacterial. every other set of jars has been alpine white, so my guess is something bacterial in the LI or grains

btw, these are all on wheat berries, and i have been using wheat berries from a seed supply store rather than baking supplies, though the grain has worked well for everything else.. makes me suspect the LI. also, none of these are intended to be grain masters or anything, just trays and monotubs.
i really appreciate your feedback my friends. my first instinct is to scrap them all and make new cultures, but figured i would ask before doing anything drastic

thats what i was thinking, definitely looks different than the last couple hundred of alpine white jars, and all the other cultures i am testing right now
should i scrap both of them now? or there any reason to watch either of them?
I suspect it might have something to do with me trying to reuse 60cc syringes by pressure cooking them, worked well the first three times but on the 4th i had a plunger fail on me. gotta check my notes to see if i used a new one for those 2

I re-use 60cc syringes more than dozens of times and don't even pressure cook them. boiling water is just fine for plastics.

my b, on my phone it looked like popcorn. winter wheat is fine. But it does look very bacteria ridden

thats what i figured :/ i wonder where the point of failure is on these? went from clean culture to LI to wheat berries, just like i have done with everything else
i usually assume at least 5-10% contam rate on anything liquid and less than 1% on jars, but i had been particularly lucky lately, and have much improved my clean tech (using lab gown with cuffs, basically all my supplies are from lab supply or medical supply places now) and much improved general technique, and have had much less than my previous tolerance for contams.
Im guessing it must be that i messed up making the LI this time somehow or another.
what do you think bodhi, should i dump out all 6 AA+ jars that were made from that culture, and all 6 PE6 that have that yellow coloration where the grains contact the glass? any reason to keep monitoring any of them, or would they be better used on testing something else?

thats a great idea bodhi, i need to get a good microscope for home, been eyeing some trinoculars so i could post pics to here, but at that price it kinda makes sense to go ahead and squeeze the trigger
i have a good scope back at the lab that i should be able to use for these but would need to prep the slides offsite, so no one knows what im workin on
in the case of the PE6, how would you go about preparing a slide for checking for bacteria/mold from a jar of grain like that?
i suppose it is probably a good idea to make a slide of the LI that i have leftover from that batch too huh?
