Colonized grain to agar

 This is probably a stupid question, but i havent read anything about it.
I have a pe grain jar fully colonized. it was made from a third transfer from agar. A small wedge, about the size of a quarter. It took about three weeks to fully colonize a quart jar.
The above pic was its recovery after a shake less than 24 hrs beforehand. I assume its pretty clean.
I was thinking if i took a few grains, put them to agar and do a few transfers to further try to clean from any possible bacteria, and get some plates with strong growth. I assume there would be no problem doing this, after all its still MS.
Then drop some agar plates to master grain jars, g2g those. Then grow out 6 to 8 mono's, and then go from there with clones etc.
But most of all my intention is to get as close as i can to bacteria free tubs. With hopefully a clean prolific tub or two.
Would this be a good strategy? Besides cloning my first ms grow?

There is no reason that this shouldn't work. All you need is one grain so its worth the try.

I do this a lot with left over grain from spawning....great leap off

Thanks, I figured that. Another question I have. Ive heard is that bacteria can intermesh with the mycelium. Is this possible to spot without a microscope? Or are there tell tale signs that bacteria is present if not picked up visually?

Thanks for this thread. i am getting ready to do the same thing. So what technique will you use to make the transfer of grain to agar. I've got a couple of ideas but if someone has a valid method i am eager to hear it. thanks in advance.

If you want the cleanest growth you take an un opened grain jar into an sab with the plate you are transferring to. Heat sterilize a pair of tweezers, and pluck one of the top grains and drop it on agar in the SAB Always practicing proper sterile technique.
Or you can do like cron said and spawn all your grain and take the extra onto agar. That been exposed to open air though and would require more clean up on agar so I don't see why you wouldn't just do the aforementioned before opening the jar up

Thank you. I had thought about it and yeah, flame sterilized tweezers is one of the things I thought of as well. Sounds good. I'll use that method.

You could drop a colonized grain on carpet pick it up with your fingers and put it on agar and it would grow fast enough to outrun contaminants then transfer and you're golden. Any precautions taken will only help
