hello all i am gonna start my first agar, tissue transfer -> bulk grain grow! Been searching the forums for days and have seen many recipes for a good agar medium.
This is what I ended up gathering up over the last few days:

I just have a couple questions: 1)Will this make a usable medium to grow? 2)Anything I should add/remove? 3)Is this method faster then say PF tek? (I've only dont PF in my experience thus far.
I will by using Pastywhyte's Agar Tek mostly. Ill use fresh tissue wedges to colonize the agar. Then transfer to grain.
This tek is all new to me so feel free to add anything that could be helpful.

Me and you in the same boat brotha. Got some plates going with pastys tek right now. Get yourself a craft knife or hobby knife. Basically a scalpel. It helps huge! I was trying to use an xacto knife before and it was frusterating to say the least. Just rip the mushroom in half and tease a little tissue from the inside. As little as possible. (Ive heard you dont even need tissue. Apparently you can just run the blade through the tissue and then the agar and that is enough, although I have yet to test this method)
Also blue food colouring!
If you can figure out a way to make the plates without condensation build up on the insides please let me know! Good luck!

That is the exact type of agar agar I have used for several years now. It works just fine. 1) yes that will work for PDA. Personally I like to use dextrose in place of Karo but have used it in the past with good results. 2) no unless you have access to a brewery store or somewhere similar and get dextrose. Other wise those 3 ingredients will do fine as PDA. 3) I wouldn't say it's faster from spores to fruit then cakes, they are different animals. Using agar could be a slower, faster, or about the same depending on a few factors. If you were to just grow from spores on agar, the plate was clean and you didn't want to start isolating on more agar, you could inoculate some grain either with wedges or LI then fruit from the grain. Not a huge yeild that way obviously, but the timing could be comparable depending on a few factors.
Agar is designed not to fruit, rather to put the mycelium in 2 deminsional plane so the cultivator can see clean growth, contamination, and sectoring easily with the naked eye.
I have never used pastyplates before, but have used 4 oz jelly jars using the no pour concept for several years now. My trick to help with condensation is to invert the jars ~2-3 hours after sterilization (they should be coagulated by that time) and continue to cool upside down. Gravity will draw much of the condensation to the lid. Upon streaking and/or inoculating, I open the jar still upside down and very quickly shake the lid onto the surface of my SAB and 90 percent of the water will pool right off. I'm sure the same can be done with pastyplates, but like I said, I have never used them personally. But the concept is the same.
