How much spores to use

Im coming back from an absence of a while and am trying out WBS and I remember from pf tek you really don't need to use more than a drop per the 4 holes but every WBS tek I read just says to inoculate doesn't say how many CC's to use or just a drop, I am wondering how much I would be able to get out of a spore syringe I also want to make an LC so I am trying to figure out how much I will need per jar to save for LC which I am torn between Karo and Sugar in the Raw ( due to I already have packets of it) any advice on that as well? Thank You

A few drops for a quart grain jar. Up to one CC
Making a LC with spores is throwing away spores. Make a LC from a clean culture like an agar wedge. Adding your spores to LC only adds a step and time to the process. Just use spores and skip waiting for the LC. Your grow produces more spores anyway...

the more spore solution u use, the faster the grains colonize.
Also, if u want a clean, efficient culture, do spores to agar, then transfer wedges to more agar, then do an agar slice to grain.

This isn't true in most/all cases.

.5cc is all you need for a well shaken syringe.
Spore to lc can work if your doing outdoor beds and you used "trusted" spore.

If you're using a syringe with nicely distributed spores you really can't use too little.
The only exceptions I can think of is old spores where you need quantity to find something viable or grain that's a little on the dry side that then relies on moisture from the syringe to germinate.
If you diluted a fresh syringe I'm sure you could inoculate 10000 jars from one 10cc syringe quite easily.
I had a spore syringe given to my recently and I used only a fraction of a drop, pushing the plunger just enough for a tiny bead to form then touched it on agar, using only micro litres each time. After doing this 8 times the plunger was visibly no further down the syringe.
If you dump 1CC into a jar you might get a little faster colonization as there are more inoculation points, but the chances of a cotam getting in also goes up with it so the extra day or two you save are not worth it.
With spores less is more.

so spore to LC to WBS wouldn't work?
Bodhisatta Im starting to read your agar tek
I also found a little article on here about distilled water I may have to take a peak into as well

Spores to LC to grain jars works but it makes no sense, A LC is more likely to go to shit than just inoculating jars directly with spores. If you need to make your spore syringe go further then just use less in each jar.
Id get some agar going before going all out with tons of jars though.

Ya, going from agar culture to grain jars is a much better path than LC to grain.

Ok thanks! I put just enough in each jar to be able to see the plunger move, Its only been 3 days and I saw one jar with some mycelium growth after the second day so I'm hoping for good things. I plan on making agar I've been reading up on it and think ill use small jar with SFD and self healing injection port, I like the idea of multiple uses vs buying a ton of plastic petri. how long does agar usually last for?

Agar is the way forward, and once you get your successful grow its cloning time and you'll never (possibly never) need spores again!!

would it be as easy as placing a untorn mushroom cap on the agar and wait for it to drop its spores and wait or is the tin foil or liquid spore transfer necessary

Easier, just take a small piece of tissue from inside the mushroom and place on agar, let it grow then transfer growth to another plate to clean up your clone. If you get a good culture you can transfer again and keep in the fridge for ages or keep transferring.
Agar seems intimidating but it's so easy in reality and essential if you want to succeed in this hobby
Go for it!!

You can do a couple things. You can transfer spores from a print by touching it with a scalpel or inoculation loop and then touching the center of the agar. Or by putting a drop of spore syringe liquid in it. Or, cut a bit from the center of the stipe near the butt of the fruit, where the Myc is the most clean and aggressive and place live tissue in the plate and clone the fruit.

Also, if you're new and your posts contain the words: Humidifer, incubator, air-stone, or heater, then you need to read and UTFSE before asking people to review your setup. OR... You should be cultivating reptiles and fish, not mushrooms.

hows the contams with the stipe transfer? Any recommended teks? or just as easy as sterile technique and drop it in the middle?

Isn't the point of a forum is to ask questions bounce ideas off people at real time, have a conversation about ideas? Please if all your going to do is be rude and not have any actual knowledgable input then feel free to not have any further input on this particular thread you have been using this site 4 months I have been for two years I've done research.
