Where are your ge filters?
Hadn't added them Yet. I doubled up on micropore tape and paper towels on top, just as I did my pasty plates...
Not a good idea apparently.
So before I PC these, I'm gonna remove the tape and just stuff tight with poly as I have in the past...
Already did 8 jars with micropore filters though. Small hole. Smaller than a grain of red Millet. So nothing stickers to the tape...
Still not advised I guess.

You'd be better off with no hole at all. Pasty has a tek for that.

Link??
Also moab you should invest in some sfds. With poly it can be too loose or tight with sfds there's no guessing. The results are more constant imo and IME.

Just inoculated my first jars with agar an hour or so ago.
Don't think it went well but we will see. I found a lot of things to tighten up. Wish I had done z strain instead of the GT as I'd rather the GT go well. But it's all good maybe it'll be okay.

The truth is that brown spots on the base are an indication of vert, as well as pseudomonas bacteria. Without a microscope you won't be able to tell what it is.
Oh and yeah clones will be a bitch to get clean.
I hear ya, but nothing ventured nothing gained right?
Just took five samples from five different fruits on this cluster and put them into pasty plates.

This is my first attempt at cloning. I have knocked on wood, crossed my fingers, toes and my balls.

I'm not sure who's post I saw that on first, but whoever it was thank you. I think it's a pretty solid idea.

I've been using a rack for a while now. Was digging hard for it today.
Then remembered I used it to set my substrate on to bottom water in a mono, and didn't remove it.... I was disappointed when I didn't have it.
If you don't have a rack, I'd highly suggest it. Your plates will never touch the floor. Nor scalpel or anything else like prints etc

If you take lots of agar transfers from spores (4-10) will the mycelium start to lose vigor?
Is it better to get a clone than to make 5-10 transfers for a mono cult?
When you have a monoculture and feed it crap will it perform like crap? If you got clean spawn and feed it the same crap it is less likely to perform like crap due to the genetic diversity?

4-10 isn't enough transfers to lose vigor. Might be enough transfers to gain vigor depending on what you're working with.
