Water bath to liquify solidified agar media? Microwave works for that (as long as your vessel and cap are microwave safe) or just make enough for the amount of plates you want to pour. Just my

To keep it liquid and at pour temp. A water bath won't melt agar and if it did it would take forever. They're super nice to have tho.

Sounds like a pain in the ass. I usually pour 600ml at a time, maybe a tad on the warmer side of things but it all gets poured in one sitting.

Yeah, I'd still microwave it to a boil before putting it in the water bath. Then let it cool down to 120 to pour. Everything in the $100 range seems kinda sketchy tho. Can't really justify spending much more than that since it isn't necessary, it just would make things a lot easier.

That's why they make em for labs right. Ive never seen a lab that does agar work that didn't have a water bath besides Alan Rockefellers space, he says he pours his burning hot

I tested melting solidified agar once I believe it starts to melt again at 85C + that it almost boiling so a water bath won’t do it

Yeah, I’m just thinking you’d need a space for the bath itself and have to wipe down your media vessel before pouring. I’d rather just pour it all in one shot. That said, I typically don’t have a whole lot of irons in the fire and work in front of the hood so it makes it a bit easier.

When you guys remelt your solidified agar you have to leave the cap on still right? I thought about loosening the cap but then it would obviously probably contaminate the agar right.

I have a question guys
I have unmodified lids for my qt jars I’ve had them since I did slurry a lot a while back. Full colonization in 70-100hours so that’s that.
At what colonization times would you consider filtered lids? Right now I’m doing BLI ala Munch and I get to full in 5days with fast cultures and about 9-10 days for the slow ones.
Am I pushing my luck with unmodified lids and 2 week cooonization?

Loose lid for sure, depending on your lid type...if it’s a threaded lid, you should be fine. If you don’t loosen it, you could have an explosion. What’s your media bottle?

I would say yes depending on the temps. Ive had shit luck with plastic lids and blenderless li ive also never had a good seal for them

Yeah I wasn’t sure because I assumed a pressure build up would be a problem but I also thought loosening the lid would let dirty air in. I’m using a Pyrex 500ml media bottle like this one

Should be all good then....no need to loosen it all the way up, just loose enough so it’s not sealed. But like I said, I’d just pour it all in one shot before it had time to solidify, especially at that volume.

I’ll probably just keep pouring them all at once. I just figured it’s good to know Incase I ever make some extra or ahead of time

Fosho The only time I ever melted any was when my boss would have some extra like that...I never cared for the practice, tbh and would always do it all in one go when it was fresh out the clave.

The bath cools it down fresh out of the clave in under a half hour. And to the perfect temperature. But where the fuck do you put this extra stuff. I wouldn't bother with one for home mycology.

What do you guys think about this? I have this one pound bag of rye that I never broke up because I didn’t feel like it (last time I broke it up a little late and it caused some bruising and then had some invitro pins) Now I have these few top grains that won’t colonize they’re stuck out of the loop so to speak. You think it would be fine spawning them or just I dig those few out before spawning?
