I'm top fruiting a jar I thought was bacterial, and there's a significant amount of water pooling towards the bottom. Is this likely to get incorporated in the fruits or should I attempt to pour out? It's probably around 20-30ml of water.

Anyone else noticed pan cyans eat food coloring? I made LCs with bright blue agar from a cube plate, an oyster plate and a pan plate. All three turned blue as the liquid took on the color from the agar, but a few weeks later now they're colonised and the cube and oyster remain blue while the pan cyan has turned crystal clear.

I haven’t used food coloring with any species but I have never heard of that, very cool indeed. Any pics?

Ive noticed every mycelium eats food coloring. Food coloring is cool for keeping recipes separate but sucks for helping see what's going on on a dish

How sure are you it's water and not metabolites or bacterial soup? How long has it been there?

It's free of any off colors and while I'm sure there's some bacteria in the jar it's less than what I thought. I'm still trying to get my prep right, so these rye berries were loaded a bit too wet. The top just started forming primordia.

Ive been reading up a lot on the possibility of cloning a dried specimen. I took a big dried mushroom and tore it open in the SAB. Everything was done in a sterile environment. I put gill material in a few dishes and a couple scrappings from the inner most part of the stem.
Its been two days since they were put to agar and there has been no activity as far as contamination. Im really hoping it works, but the more i read the more i realize it may be a very difficult process.
Also, i made my first spore syringe and put it to agar. We'll see what happens. Just wanted to share that with yall.

It took a minute for me, about 2 weeks before I could transfer. I hydrated in tap water for about 10 sec then dunked in iso, then plated. I put 3 samples on one plate to increase chance of recovery. The growth looked terrible, but after I cleaned it up and allowed it to fruit, the growth from the first clone looks very nice.

I think food coloring is really good for seeing contaminants on plates. I wouldn't use colored for a LC but the contrast seems to really help.

I find transparent agar to be best for seeing anything going on. But whatever works for you is the best for you

I never let water stagnate for more than 24 hrs

You have to buy 1000 at a time with those Unicorn bags though don’t you?
I don’t have $300 to drop on bags all at once so I tend to go 30-50 at a time

I've been working through my 100 bags I ordered. Was $60 or $65 with shipping and they are sturdy. Not as bad as dropping $300 at once, but I still feel like I got a good deal at 0.65$ a bag. I only use them for sterilization of substrates (I don't do monos anymore so I never need more than a quart of grain) for edibles so they should last me a little while.
