anyone spawn too still steaming hot coco?? i completely spaced out while dumping a tote worth of spawn directly into the bucket after like 2hrs:facepalm:

Brilliantly put chron.
All I would add is there's a difference between cloning, isolating and stabilizing. This convo is a little all over, cloning isn't hard, isolation and stabilization can be. When I'm underwhelmed with ms potency I look for a better clone :shrug:

It's not ideal, but I've seen grains from used jars survive being washed & dunked in 140+ water, so...

So this cluster of pins that came from this mushie that came from this pin. So basically at this point it's just all the same genetic material and probably was after the first clone. It really doesn't matter if I grab one of these and clone it at this point if I already have cultures of this. I'm pretty sure I knw the answer to this. :smile: I just wanted to show off how I get better pinsets on agar than I do in my shoe boxes. I still can't believe all those mushies came off of one tiny leftover agar chunk. This is what I have in jars now.

Ive just gotten into agar and isolating a strain, but so far my attempts have been fruitless. I mean, ive done everything by the book and still have this damn trich that is apparently hiding in my otherwise healthy looking isolates.

I isolated one disk through at least 3 transfers and they look great and void of anything weird looking. In fact, everything works like a well oiled machine from inoculation with MS syringe till i put to a coir bulk substrate.

As soon as my cultures go from the agar dish to the bulk trays, trich appears within a couple days. Ive lost 13 tubs in the last 4 months with not one success.

So heres what i did to at least try to get a clone specimen. I took each individual spawn jar and instead of spawnibg to bulk, ive mixed each jar with equal parts of jiffy seed starting mix peat/verm/perlite/ lime and then put about a half inch of the mix on top as a casing.

So far, there is no contamination, only mycelium. This is starting to make me think my contamination is coming from my bulk coir/coffee/verm/ gypsum mix, instead of my agar cultures.

Either way im starting from scratch with fresh materials. The MS syringes i used were 4 years old, but was in the fridge that entire 4 years and i didnt think it would be a problem.

Yes, you are correct sir. I just hear so many trusted cultivators like RR saying to add coffee so i guess i was just taking bad advice or outdated advice.

I mean, the grains of coffee in the substrate are pasteurized at 155 degrees in mason jars for 2 hours. So, i thought it would be fine

The syringes definitly must be replaced with fresh ones. This i believe is the source of contamination. Ive gone over and over my sterile and pasteurization techniques and it baffles me as to how anything would be surviving pressure cooking and pasteurization.


Stop putting coffee in your substrate & get some new syringes.

Get new syringes is a good advice, but you can try to get something of your old ones on agar just for fun and practice.

This culture has been doing me so well I am considering setting up my 90 or 105 qt mono........ but alas, who doth need that much APE :hamletmonkey:

I am trying the same clone plate in a shoebox (4 days in).
I also cloned a pin from that clone plate (which already was a pin clone) that had a sexy blue head and that culture just went to 2 qt of grains to replace this culture in the bootbox.

I'm loving this so much, what a beautiful variety! I also put MS to agar. Between these babies and trying for golden halo, I'm having fun.

Clone question:







So strange...all three are from the same fruit.  C1 and C2 appear to be monocultures, with uniform linear mycelium...then C3 is faster growing and rhizomorphic, growing in a slight spiral, and may be sectoring.  I figured they would all be the same at least.  Anyone know why C3 would be so different?  Bacteria?
