If working on a culture that is bacterial would it be wise to take a transfer growing up the sides of a plate.

My thoughts are that a dish has still air so if anything bacteria  wise is in the dish it's going to struggle to go up the sides of the dish

Let alone there is no nutes on the walls for growth.

What do you think.

Thats interesting.

This is my old culture when im new. Most of my Dfagar crawls to the sides.
Im not sure if my memory serves me right that the side growth transfers did produce invitro pins on some dish.
Hope i can recreate this to test for side growth transfer.

I can sense that im facing this problem again since im getting 2 kinds for growth (aerial and flat) 50% from this

Donor dish. (4th transfer from spores)

Aerial growth

Flat but some aerial radiating from the center.

This is biopsy from the donor dish.
As you can see theres aerial growth from the top, then some rhizo clean strands from the bottom of the dish.

Im Still afraid to noc some grains from them prematurely, since ive experienced this before the bacteria did bother the mycleium resulting in slower colonization.

I did so.e hot pour from the donors for test.
Hope some can chime in.

So the guy only has these as replacements. Can I just get a variable speed control if the flow is too heavy?

TBC- I see nothing wrong with that filter. Just make sure your plenum is at least 9 inches

Oh it is for sure. I have two of these units already but they were broken in the mail so these are the replacements. My ex fixed one for me. The other one is physically beat up but works fine. But only if you put it on a certain side. If you flip it any other way it makes a sound I don't approve of. I vacuumed them both out and got some of the larger particles out including a few flies. Eeewe. Looks like they never had prefilters installed on these units. They weren't that bad besides the flies though. Filters look like they still got a good year in them at least. I also just ordered a 6 pack of Merv 11 1" 16x16" filters from discountfilters.com for around 37 bucks which is a fucking steal compared to the almost 20 dollar price tag at the stores. You can pick them up with the code deal20 for the extra 20 percent off and the shipping is free so I saved a shit ton of money. I also bought an analog humidifier for my Inkbird controller I have coming in this Sunday. The humidifier I have now is digital and uses a touch pad. Fancier is not always better. It served me well for several months. And I got my bags of lime coming in so I can pasteurize this damn straw. I say I pretty much have everything I need for a long time now.

I'm going to build the Great Wall of Laminar! I have a local guy who may buy one of them off of me but I'm not sure I want to sell it. They are so expensive and who knows what I will be doing with my mycology in the future.

Can anyone recommend any good well fitting glass petri dishes that can be put in the PC? I recently bought some, and the tops were way bigger than the bottoms. That was frustrating!

I haven't really heard anyone ever recommend glass petris.  They are apparently a real pain in the ass to work with due to how slippery they are.

You could just pasteurize your straw in the stove like I did tbc, it only takes forever lol

I've also heard that the process of cleaning them and sterilizing is more costly and has a bigger environmental impact in the long run. Plus they aren't cheap, I'd hate to drop one or knock over the stack when pouring. Anyone who has dropped a hot pipe can tell you how easily borosilicate can break when hot, and pipes are way thicker.

I do well using these jars with modified lids. I just mix the agar. Pour it in the jar and put a plastic ball lid with a quarter inch hole and a synthetic filter disc piece cut out to fit glued on with RTV. Been using this method forever with really good success.

Man, I fucking hate WBS. The shit gets everywhere and no matter what I do, the fucking things wind up being sticky as fuck. I'm assuming it's starches coming out the shit, blah. I'm thinking I should rinse them off in cold water before loading so they don't clump in the PC.

How do you prep them.  Yes the millet gets fucking everywhere hard as hell to pick up.

I just used hamloaf's method which doesn't include a cold rinse after. My WBS probably has way more corn than his or something. Going to rinse it all and load it wet.

Yeah, fooman's is what I always used before and then switched to oats for the last 2 years or so. I'm going to go buy some more tomorrow, fuck this millet.

Are you prepping with heat? Try just soaking it, that’s what I do.

Fooman’s prep always ends up with clumpy and overly wet grains for me

:whathesaid:

I never had success with cooking wbs so I only soak it overnight. No cook wbs all the way. I rinse it after and spread it out to dry and try to never load it wet (especially if I'm pouring an Li or Lc into them later)

I'm using small budgie seed, but I hydrate it differently. I add boiling hot water to my seed, and leave to cool for ~1 hour.
I then strain and weigh it, and repeat if needed, until 60% water, then I assemble it and pressure cook it.

Cooling to room temperature causes the seeds to absorb water, and not burst or leak as much.
WBS can also contain naked grains and other, which can also breakdown easily.

Well I decided I’m gonna start up a little greenhouse. I only want to make one that’s like four feet tall though as I’ll be moving in the next couple months and don’t want something big and bulky to move.

I figure a four tier shelf with a martha over it and cut the bottom so theres a good 8-12” exposed for FAE? Or is using a little fan a better idea?
