Do you have bricks or loose coir? If you have bricks hydrate with 4 qts and add verm until at field capacity. Spawn at a 1:2 ratio. Whatever you have left over lay out on a baking sheet with a fan running next to it, mix every few hours until dry. Then you can store it for later use

Maybe 300 g and 2 quarts? Add 1/2 qt water for every quart vermiculite if you're using any.

This sounds like a fair ratio, basically about half of what a brick of coir calls for, should make for ~4 qts of sub to split betweeb two boxes

Depends how big his shoeboxes are, but mine are 6 quart and I make 4-5 from 1 brick of coir, depends how much spawn is used too obviously.
2.5 quarts water for half a brick coir and 1/2 qts verm.

Yeah it keeps for a good while, a had some leftovers in my bucket for a month and it never greened on me. It just dried out.
You'll find it waaay cheaper online and you can order a case of it.

[Personal preference]
I prefer to work with weight, as volume does not tell you how much it weighs in total. For example 100ml of coir might weigh 60g, 100ml of water is ~100g.
Take a look at my substrate water content guide below.

I make half a brick much more often than a whole one. Probably because I mostly grow in shoeboxes. To each their own but I’ve noticed a difference between using freshly cooked coir and coir thats sat in a bucket in a closet for weeks, whether its because of added water content from accumulated condensation or because of an increase in microbial life in the coir that the mycelium takes longer to colonize, I don’t know. But after a year of bucket tekking and making shoeboxes, half a brick is my routine

Cool, as mentioned it's also my preference. I found hydrating small amounts and then adjusting easiest. There is some overlap based on cooking time, where a larger amount would hold more water.
I do capacity I want + [X] amount for cooking losses, to get nearly exact. You can measure the start and end weight of a test run.
You end up with [X] water for [Y] minutes.

Does anyone have experience cloning mycelium that's expanding in the substrate?

I have a shoebox with a few patches of extremely aggressive mycelium and I'm strongly considering slicing off pieces to put on agar, clean it if needed, then xfer to jars of oats to expand via G2G.
Thoughts/suggestions?
Btw I have gentamicin if I need it.

Yes I'm sure. I have more than a few boxes going and haven't seen trich since I started back up a few months ago. The picture isn't super clear I know...

I just triple checked, it's eatting the coir and not affecting the surrounding mycelium. The one patch is pretty big to not have sporulated yet also, ime. If I isolate a super contam I won't say you didn't warn me.

i definitely should have bought bricks instead of a giant block that i have to hack pieces of off. bricks seem to be an easy unit of measure around here. definately reading your substrate water content guide.

At the top of each post a user makes, there is a "+" box on the end, to the right. Similar to "like", it's "vote up".
----
You can also use my guide for grains, weigh the dry grain, lets say rye grain, the maths:
100g - 10% = 90g dry (difference 10) | 90 x 1.28 = 115 | 115 - 10 = 105g water. 205g total, 115g water / 205g total, x 100 = 56.1% water content.
100g - 10% = 90g dry (difference 10) | 90 x 1.50 = 135 | 135 - 10 = 125g water. 225g total, 135g water / 225g total, x 100 = 60.0% water content.
Measure the end weight of the setup, and measure losses.
