here is the way i have been streaking spores, seems to really cut back on the number of strains present in my initial transfers, and gives me lots of options, really helpful for isolation

Spores are quite larger than bacteria you might only really need to go to step 3-4

I don't have an inoculation loop. Been trying to fill my Amazon cart for a while now... Found 5 packs of telephone agar for 10 bucks. An inoculation loop for 6 bucks, 200 sterile swabs for 7 bucks.... added some other shit in to make 50 for free shipping.

Maybe I should order some petris while I'm at it.

You have to see what those classify for free shipping i cant remember if it was 50 or 75 but still a good deal i fucked up and ordered my pc before seeing those i tried getting 3 and shipping was 41 haha so i will just be getting wally world jars haha

Unless working in front of flow, I would much rather not have my plate open for so long.  quick swipe in the center of the plate with a spore loaded loop or scalpel then shut her down.  If you take a small transfer on the first transfer, you will quickly get plenty of sectoring to test.

But for me, I usually just transfer a few times til nice and clean (selecting fastest/most agressive sector areas), and then grab a really nice cluster clone from first grow.  that is how I have gotten all of my best cultures.

lol thanks guys!! i love that gif, that technique saves so much time. i had been doing it incorrectly forever, i would scrape up spores for each step lol....

you dont have prime? amazon prime rocks!! i love that shit. especially if you know how to get review products for free :cool:

id add some things to that list: pp5 test tubes with top, box of 100 sterile scalpel blades, at least 4 scalpel handles, a big roll of parafilm, 500 case of petri dishes, that red 4oz bag of agar agar, media bottles, glass stir rod with rubber policeman, set of food coloring powders, etc.... you know, just everyday shit for a regular human

You can use the simple soak method for all grains if your just looking for spawn.

I got a quick question if anyone have a good answer I ran across this guy who been doing grows for awhile he told me once I have a fully colonized monotub I am able to pull a core from the tub and transfer to another one to colonize that one a 2 for 1 type deal is this a reality? Mostly asking cause got 6 jars about ready to transfer to bulk and was hoping to case one and just fruit other right out the gate as experiment.

I wouldnt try it i mean what are you going to do with the chunk you cut out and you would have to literally take half out and break everything up and re colonize allowing the mold and other nasties to get a foothold i woukdnt try it

Ya he said that you just replace the core u take out with a core from the new manure. He recommend use just a sterile copper pipe but with all that exposure to outer air will increase my risk to contm. correct? Thanks 4 info

this guy you ran across doesn't know wtf he's talking about. You can do something like that for an outdoor grow, but not indoors.

You can do grain to grain (g2g) transfers to expand your spawn. Each quart of spawn could be expanded to as many as 10 more quarts of sterilized grain. With clean spawn, this is how you'd do that for indoors. That's spawn; part of the substrate, not a core sample from a fruiting tub.

Or, you could take a clone. Peel apart a fruit in front of a SAB or flowhood, take a biopsy and put it to agar. This will grow out, and you can clean up your sample, and inoculate more grain jars. Sounds like this guy had a couple of things confused maybe.
