Ehhh been kinda scatterbrained lately. Those agar circles just pop out don't they....
1.6,2.5,3.3 glucose agar potato is the recipe I used. To half a cup of water. Via the pasty plates video Tek. I tried to just follow that. But I think my scale is off by a point and I didn't use Karo I weighed out 1.6 or so of Agave Nectar
How can I make the recipe better? Boil potatoes instead of instant flakes, and just a drop of agave nectar?

Munches and my recipes are pretty rich to begin with, idea being most new people are gonna be germinating spores and might benefit from a little extra nutes. But adding nutes on top of that is gonna be detrimental in the long run. 1g sugar and 3 g flakes to 120 ml water is like 3.33% nutes. That's plenty. But up those to 1.6 sugar and 3.3 suddenly you are in 4.1% nutes. That's a bit much for cloning, too much edge given to bacteria with all that sugar.

So your verdict on the perfect numbers for a noob using one half cup of water starting with spores to agar would be? If you'd be so kind please and thanks. Ready to try again. Got in an excited frenzy last time.
1 gram sugar, 3 grams potato flakes, and 2.5 agar?
Bear with me. Sorry. So I think I should buy Karo regular corn syrup yes and use that as my "sugar"?

I have used agave. It's not perfect but it's not the reason your plates contamed either. Karo is good, or SITR. I don't like honey myself but it will work too. Don't over think this, just follow the tek precisisly until you understand why you are doing what you are doing.

Maybe this clone plate will be ok. Finally got a decent photo. If you look at the chunk of tissue, there is white myc looking fuzz finally. I assume the pretty streaks are contam yes?

That looks pretty bacterial from here to me though.. Isolating away from that could be a pain. How was the clone sample taken?

Not very great I don't think. I took a fresh mushroom off a PF cake, put in sab, ripped open, took a chunk of Stipe from the insides with tweezers and placed into upside down opened plate. I had a hell of a time getting the chunk off the tweezers. I think I swiped,tapped, rolled... it took great effort to get the chunk off the tweezers. I think maybe I flung a bunch of bad air around or something.

The reason why we ask this question is it is very common to get a bacterial contam if you use not the inside of a fruit body but just a piece of the stem.

Anything that bacterial suggest a procedure vector. Use a scalpel and just tease off some flesh from the inner stipe. Caps are harder to pull clean tissue from. Tweezers are a pretty ham fisted tool for tissue collection IMO. Use a scalpel and it's almost a minor scraping motion. Only need a tiny tiny amount of tissue. More means more room for bacteria to hitch a ride.

The hardest part with cultivation is sticking to the script exactly. Sometimes that means going a week without doing anything because you just don't have the PERFECT setup yet. It's worth the wait, always.
