go straight to grain in a half pint or pint jar only 1/4 full. then put some grains to agar once it germinates. or if it looks clean enough g2g it to a couple quarts and if those look clean g2g those out and then grow and clone.

This is basically my new favourite way to germinate stubborn spores. I'm having a bitch of a time getting spores to germinate on agar these days. No clue why. I do know I'd much rather work with living myc, so get them to germ on basically grain plates, then move that to agar to clean up or like bod said, g2g the shit out of it. OR, spawn it to something really small and grab the first pin you see.

If a nice set of genes do pop up on the first flush, rather than hastily grabbing the first pin that's seen, let the fruits mature, and inner tissue sample the fruits that display genetic characteristics that you would prefer to carry on.

Yea that'll probably be my next step then. I'll throw a few pint jars in my next grain run and see if I have any luck.

I g2g two different clone master jars and cloned a fat atl#7. I've yet to put to fruit cloned culture. Im excited to see the results. I've tried in the past to fruit clones, but all ran into issues and became contaminated.

I had pin growing from a 1 of my first accidents a pin i made it grow in like a s by putting on top of the lamp, i think direct light gives it a stronger direction reflex to light to face when growing. Thats my observation

I have 2 6500k bulbs in the ceiling light, and one in a floor lamp that's about 3-4 ft up from the tubs, plus some ambient daylight from the window. They also get about 20 mins of direct sunlight every day as the sun sets.
I think as long as they're not in complete darkness, they'll grow fine. My buddy grew some in his closet, from cake to fruit, in complete darkness and they were fucked looking, but he had a crazy harvest and they still got us fucked up.
I'd still keep the light.

Explicitdetail said: So I'm having some trouble germinating APE on agar. Over the past 6 weeks I've failed to get any growth on MEA, PDA, or Grain Soak Agar. My temps are around 74 ambient light 12/12 and still haven't had any luck. I got the syringe from a sponsor so I have no idea the age but I'm thinking either force hydrating or putting them straight to grain. Anyone have some suggestions or tips I would greatly appreciate it
I had issues germinating ape spores from a Vendor syringe a while ago myself. Out of frustration I completely saturated a plate with spore solution and ended up with five or six spots of clean growth to choose from.
I'm not saying this is a reccomended method of germinating from a syringe but I will say that vendor syringes are pretty damn clean and its worth a shot. What do you have to lose other than a plate and a couple squirts of solution?
Make sure you give your syringe a good shake before using it.
Josex also has an great method for germinating stubborn spores where he whips up a batch of BRF cake mix and makes no pour plates with em. After the spores germinate he moves the myc to agar and cleans per usual

Honestly I did the same when I started my last batch from syringe. The saturated plate was the only one to take off

So new to PE...How common is it for PE to throw regular cubes in subsequent flushes?
Mine are doing this and producing a lot of spores before the caps open fully..
