Yeah, that's quite a bit of bruising though. More than I've ever seen on cakes. You been slapping them around?
Most likely from drying out though. Perhaps rough handling or too much fresh air. I wouldn't fuck with it too much since those cakes probably don't have much life left in them. One on the top right is all aborts.

Hey guys, I've been wanting to start using bags. Is it a pain to work with in the sab, should I get an impulse sealer or does folding and tape work fine for spawn bags? Anything to avoid when buying syringes for liquid culture? Thanks for any tips or info.

I use a vacuum sealer on .5 micron bags and inject through the bag with LC and then put duct tape over the hole as soon as I pull the needle out in the SAB I know people have opened bags in the SAB with success but to me that sounds like stuff that should be done with a flowhood

You should assume that any LC is contaminated and verify it's cleanliness on agar before using it. Just because you paid someone for it doesn't mean that it's clean. If you're just going to inject LC it's not a big deal to use a SAB as you can leave the bag rolled up and just inject through the side - If you want to G2G to bags, a hood or FFU is pretty much required to be consistently successful.
Myers Mushrooms has good pricing on partial/full cases. T series bags are .2u filters - just pick the size that you want to run. 3T's are commonly used with 3-5lbs of spawn for tubs

You don't need an impulse sealer, they are VERY convenient but have their disadvantages too. You can get away with folding the top down a couple times and add a spring clamp or two. Some people use tape also. You could do your SAB work with tape/clamps then seal later if you really wanted. For LC syringes, I'd recommend larger gauge needles, syringe size is up to your needs.

Thanks everyone for your input. I'll be making my own lc for the first time and I'll definitely check it out on agar first.👌 lc should be easy, it's just getting that first hand experience.

So it’s been 11 days since I nocc’d up some WBS agar with Texas yellow cap. 2 prints, one labeled “dirty”…which I can attest haha, all but one of twelve have bacteria and/or some pink shit. The 12 from the second print are all nice and contam free.
But the dirty plates seem to have taken off a lot faster whereas the clean plates are still looking kinda weak in comparison -all the plates from both prints are tomenstose at this point. (this is my first go and I’m not sure how myc should look at this stage)
My question is this, What would you guys do?? Make some T1’s from the thick but dirty plates or let the clean ones ride out longer and T1 from there…or both?

I have to say that even though this is my first go, I did plenty of research and test runs beforehand, including intentionally contaminating agar so I knew what to look for. The ones with “no white”, actually do have white, it’s just very very thin/fine (that sheen on top was first indication of germination) and presented on all the plates prior to noticeable fuzz. The contaminated plates actually presented their contams days before any signs of myc. My gut is telling me that my wbs agar recipe may have something to do with the fuzzy tomentose myc., or like I said, just don’t know how myc behaves this young.

That shine/sheen is bacteria. Likely slime bacteria or something idk. Regardless it’s bacteria and I get it too.

Personally I don’t like any of those cups. I think you could attempt water agar to outrun the bacteria but if there’s mold you’d still have ride alongs. Let someone more experienced chime in. At the very least you’re gonna need a few more transfer.
