CBK, if you try it and it happens to work out, that'll give me hope for a project I'm trying to attempt, and if I'm able to pull it out it would be the most rewarding thing I'd have done so far.
My situation is: -Swabbed dirty af APE swab on a lot of plates some time ago. -Only got germination in one plate, but as things developed I'm almost possitive now that the growth I saw was not from spores, but from a piece of tissue on the swab. -I took 10 transfers from that plate to blanks. Growth was weak, slow and it carried bacteria with it, so I cleaned it a little more by doing another transfer. -2nd gen plates still look a lil bit off, so I let them grow out till 80℅ colonization of the plate, hoping myc could overtake bacteria. -From those plates, I stabbed the cleanest spots with a syringe and transfer virtually microscopic pieces of tissue to some pf tek plates. The idea behind this is that I think bacteria will have a hard time to expand quickly on pf tek sub and sure enough I got what it looked to be clean growth. -Transfered from those pf tek to more blanks pf tek plates using the same syringe method and used these last plates to make LC. -I plan on fruiting these in shoeboxes but I'm not expecting great things from it, as I'm sure what I have here is weak and slow growth from a clone tissue. -And finally, if I happen to grow one single fruit, I'll use that trick I posted to get some spores and start fresh from MS.

Btw, forgot to mention... I tried to buy an APE spore syringe from sporeworks, but the system didn't allow me to submit my order.
I contacted them several times via email 2 months ago but they didn't answer back, I guess that's their nice way of telling me that it can't be done.
Is there a way I can contact them here on the Shroomery?

When you look up shroomery sponsors there's usually a username beside the sponsor that you can message on the shroomer directly

Hey guys, up until recently I've never had any bacterial problems while working with agar.. none I couldn't fix on my own.
I've recieved quite a few prints I've been trying to germinate and I just can't seem to get them to germinate without ugly bacteria beating them to the punch at the innoc point. I've read that bacterial problems are normally user error, however I only have problems with some of the prints I have.
Any pointers? Tips? I'm fighting to have these cultures and I feel I'm at a dead end. 3 separate agar batches have been a dud. Thanks mushcult.

anyone got a great spore syringe agar germination recipe? inoculate two APE (on a plate!) pasty plates 10 days ago, not seeing anything....
I often am not the best with getting plates to germinate. I can keep them going once they start, but getting them going is usually a task for me, for some reason.

The best way to germinate spores from syringe is to prep some 1/4 pint grain jars about 1/4 full of grain and inoculate. Then wait for some good growth and snag a colonized grain from the top and drop it on agar

That's original as hell. I have a half pint just like the one pictured, 3/4ths full, already pressure cooked, but un inoculated.
Drop some APE sperm water in that jar and wait?!

I'd dump out some grain before inoculating though. You get a cleaner culture the quicker you can snag a top grain, kinda just like taking the leading edge of myc on a plate

You could get good results going to agar...I'm pretty sure I've got a few monos from spore syringes lately...Spore dilution can give you good quick cultures. Correct me if I'm wrong please.

I've germinated syringes fine on agar but I've just found that you get cleaner growth very fast this way. And those stubborn spores that don't like to germinate on agar seem to always germ on grains

I'm going to try this tonight for my Ovoid, they hate me. 3 weeks of seeing spore clumps sitting on agar not doing shit!
