Ahhh, I got cha. Yeah, that's gonna take some serious research. Hopefully one day we'll figure it out.

Dendrocopos said: Maybe a bit of topic, but I really like a comment i saw from someone on here, maybe even in this thread, it was so accurate. It was a response to a guy who had mysterious issues with all his tubs going green even though he had good cultures. It went something along the lines of.
"First will come the mycotoxin crew and blame it on chemicals. Then the mycoparasite people will stick their head in, followed by the believers of left over starch in your coir prep bucket causing problems."
I dunno, feel like it sums up the shroomery really well Not saying any of those things aren't true. Accept maybe the starch thing i find a little bs (unless you have huge amounts of visible starch in the bucket), altough i have dedicated buckets. Just, the only person who can help you is yourself, cause everyone has specific problems and materials. The best advice to give is to get a notebook and do experiments i feel like
Some mycelia can simply turn to shit overnight (or, more precisely, over 1 transfer). Had it happen with my most beautiful and uniform Pan Cyan culture. One transfer away it barely grew and was all kinds of fucked. No signs of contam anywhere either, smae agar prep as always and everything. It just went full derp-mode and never recovered. I wouldn't be surprised if that's been an instance of a myco-virus, but then again it could have still been many other unknown factors.

Yeah for sure. I have had that happen. For me, the biggest mystery is what the fuck those scalloped plates are you get from Ms sometimes. If you know what I mean. I am sure that is some viral shit too.

Any one have advice on how to get strands of swab off a scalpel onto agar smoothly?
It usually takes me quite a bit of roughing up the agar to get the entangled swab off the scalpel. Sometimes I need to get a loop involved.
Maybe I should just break the wooden tip off and use a long tweezer to jam the cotton into the agar.
I really don't want to waste cultures sent to me.
Thanks in advance!

Sterilized tweezers are nice but you usually still need to rough up the swab with a scalpel. Honestly though, I just iso my hands really well, grab the stick about an in below the swab and jam it in the agar. I haven't had any contam issues by doing it that way so far.

You can scrape the spores off the swab without any fibres by doing a quick swipe with the scaple. It will pick up plenty of spores. I did this recently to put spores on a microscope slide. Definitely got spores and no fibres.
Edit:ha ha, it was ^^ Bigfoots spores ^^ ! Nice and clean too.

When I get strands on the tip of the scalpel I usually cut down at a slight angle into the agar and then kind of drag the blade out so the strands roll off. It’s hard to explain but the main thing is to make sure the fibers don’t wrap up around the blade as you press them into the plate.

Actually realy good tip, i have that problem sometimes when taking tissue from clones.

The colder the scalpel the less sticky towards tissue. Also i am pretty sure that if you just scrape for a bit and slice through the agar, you'll get some tissue, even though it won't necessarily be visible to the naked eye.

I'm having good luck at 1 to 7.5
the recommended 1 to 5 is insanely dry for that brand of coco.
Fuck I’m confused, pardon my low IQ.. So for 1:7.5, for every lb of coir I should add 7.5 lb of water?? That’s like 4L per pound of coir! For a 10 lb brick of coir that would be 40L. That can’t be right, what am o doing wrong? How many liters of water go into a 10lb brick of im going for 1:7.5?

Using bods bucket tek I'm getting 15.8 liters of water per 10lbs of coir.
10lbs = 4535 grams 4535 x 3.5= 15872
You can add more water to get to field capacity or add verm/ wring out coir if it's too wet.
I always shoot for less water.

Thanks Bigfoot! Mr.blue & AKA for the swab halp!

A big thing that I’ve learned, is that once you get confident in your ability to produce clean spawn, using much more water makes a big difference in overall yields. Sure, when starting out people are told to make it too dry vs too wet, but that’s because of using less than ideal spawn. I use about 7.5 gal of water to one 5 kg coir brick and about 4 gal of verm nowadays and I have noticed a significant increase in yields. Mushrooms need water so don’t be afraid to give it to them

I agree, but it is easy enough to start on the drier side and then increase the moisture level after casing and get the benefits of a drier colonization run and a wetter fruiting phase.

Sure I guess it’s kinda easy to be constantly manipulating the moisture content as a tub colonizes and fruits, but I’d rather just make it wet enough at the beginning so I don’t have to touch it till it’s ready to harvest. I think that’s easier
