I need to sterilize my scalpels and loops today to do some inoculations and transfers as well as make some agar to pour some new plates but I won't be able to pour before the bottles solidify and I would prefer to not run my PC twice. Backing the lid off the media bottle to microwave makes me nervous because it sucks air in and I have cracked media bottles in the microwave by not backing the lid off. I always just pour before the agar solidifies but I did a poor job of planning my workflow so far this year...stupid LAGM lol
Microwave for 5 minutes. It probably won't go the full 5 minutes so don't take your eye off of it. As soon as you see it starting to boil at the top, take it out, tighten the lid more if you can and let it sit. A few weeks back, I nuked it for too long and the cap loosened itself. Agar sprayed all over the kitchen counter lol.
When I had covid and couldn't leave my room, I used my sousvide. I set the temp for 197°F and it took like 4 hours to fully melt. The sous-vide is nice. I always use it for agar. Microwave the bottle, let it cool for a while and put it in a sous-vide bath set at 115°F. It'll stay at that temp as long as it's running. It's nice to not have to hover over the bottle constantly checking and waiting for the agar to hit pouring temps.

One thing for sure the center takes a lot longer to melt than you expect. Damn things!

My microwave lets you change the power level between 1 to 10 so I just set it to power level 2 for 20 minutes and use that time to prep the plates and work area for pouring. Requires a bit of dialing in but I don’t have to worry about boiling over in the microwave anymore.

I know you said that you didn't want to resterilize, however the last couple of rounds, I used my Instant Pot.
- Pull the solidified media bottles from the fridge the night before
- Place them in the Instant Pot with 2 inches of water and with lid and vent closed
- Set the timer to start about two hours before I get home from work
- run time of 5 minutes because it is already sterile
- Keep Warm setting on low to run for several hours after that
- Get home to perfect pouring temperature bottles ready to go

I typically take a piece of styrofoam, cut it into a rectangle that fits snug inside the smallest priority mail box which typically cost around 8 bucks to ship, then cut a circle the size of a petri out of the center of the styrofoam rectangle.
Fit the petri into the hole then slide the styrofoam rectangle into the priority box.
I've mailed multiple petris this way with no issues.

OP on this site used to mail me prio shipping boxes the small ones that u can cram 6 petri dishes into. I only asked for 3 he sent 6 anyway lmfao. They showed up fine. Only did it once tho. Probably wouldnt super risk it but what tf is they gunna do? Fruit the damn myc to see if its an illegal culture?

Yeah, couldn’t they just be mislabeled as oyster. They mycelium on a plate surely doesn’t contain enough illegal chemicals to be easily tested.

All I know is my buddy just handed me plates of some of the harder to get things. They are fucking blue on plates and pinning like wild. Enigma, tat, those like.
I'd say a small small fucking wedge in a dimebag.

They would need reasonable cause to open any cultures for testing. So you would have to be on the radar and they intercept you kind of thing to get caught or obviously label it. Nobody is doing dna test on random cultures in the mail.

So there's no active shit in mycelium? I always heard the opposite. I was just reading this biology book and they were highlighting the beneficial compounds tested on brown rice flour mycelium, it even had me reconsidering the whole LC as edible concept idk. But what I'm wondering now is if you dehydrated mycelium and tested it with Elrich? What do you guys think? I could try it right now
Has anyone ever tested mushroom powder with Elrich??**

So I decided to take my media bottles from the PC and put them into my crockpot. My tap water comes out at 140F so I filled the pot about half way, put the media bottles in and set it to 'keep warm'. I just checked after ~2 hours while working in the SAB and the agar hasn't gelled up at all and the water is reading 117F so it's at perfect pouring temp. This may become my go to method

@bigfoot I bought a cheap ass slowcooker from a corporate giant department store for $19. Its 5L and holds three 1L media bottles. I use this to keep my agar liquid when I got heaps to do.
Regarding mailing cultures international, I have received a slant tube of herecium erinaceous from the US that made it through customs without being checked. I have also had small agar slivers in dimebags sent in the mail from the UK. Hell, I even had a friend mail me a gram of blow in a birthday card from the US once and I have seen plenty of molly come in from Europe that way too. But I have also had several letters with prints go missing/possibly intercepted. I think it all depends on whether you are lucky/unlucky enough to get the random mail check. I do know they have a zero tolerance for obvious seeds, spores, cultures etc here, regardless of whether it isd labelled or not. And now they have just brought in a law that we have to sign a customs declaration for anything leaving the country that isn't a plain letter. That way you can be traced and charged with an extra offence ie. lying on a customs declaration as well as the customs violation itself.
I hear that in AU they randomly check approx 3% of mail. I would bet that number is much lower, just due to manpower and sheer volume. Most of the big drug bulk comes here in sea containers, of which they say they check 1%... I find it funny how they're more concerned about stopping postal violations instead of the big guys hauling bulk

OK so I was wrong...I need to set it to 'low' not 'keep warm' I just poured an entire sleeve of chunky ass plates lol at least the first sleeve is mostly smooth pours..As long as they're clean they'll still be good for something. I guess that's what happens when I've got too much to do and not enough time.

The crockpot idea sounds good. Let us know how the low function works. On low, mine gets pretty hot.
This is what I use. It can only hold one 500mL bottle at a time, but I never pour more than 20 plates at one time so it works for me. IVYX Scientific Water Bath:

I’d try to find it used. It works well, but I don’t think the convenience is worth the full price, but you can leave your bottle at the perfect temp for long periods of time. It will shutoff after 2 hours of 60 degrees Celsius or above, but mine’s never high enough to trigger the auto shutoff. I set it for 50 Celsius and it stays at about 48.
You can easily accomplish the same thing for free at home, but I like being able to PC my agar in the morning and then pour at night. This makes it so I can leave for hours, come back later and it’s ready to go.

God, flow hood didn't fix bags for me. Why 😭
1. Made bag 2. 5d on shelf 3. Transferred a kernel through a cut, taped the cut(not porous) 4. All bags contam

Bag gets folded put in PC. Left on shelf forever. Bag gets removed from shelf wiped with iso. Opened in front of hood I dump grain in. Then seal bag.
The bag is sealed and sterile till you open it. No need to seal till inoculated IMHO.
I'll post this till I get sponsored...

Bags are large masses that create large turbulence in the flow hood.
Don't crowd the hood for one.
Make sure you keep your hands as much out of the way as possible when you open the top and pull the bag open.
Personally I elevate the bags off the work surface with a rack to keep off the turbulent table as well, so nothing rolls up the backside.

Why are you cutting into the bag to introduce a kernel?
There's no way to do that unless you work sideways in a flow?

I gave it a good mist and changed out the vents with filters. Seems like it responded well but not sure what is happening here. Would appreciate any suggestions or feedback.
