when I do blended LI I see the most growth where water pools at the bottom of the jars not where all the tiny bits of agar are dispersed throughout suggesting that there's tons of tiny little strands of myc suspended in the water that you cant see. I bet theres more myc in the water than you realize but well see. did you squirt the agar out of the syringe?

That’s what I’m hoping for.
No the plate was pretty hard so I just used the water and left the wedge in the barrel.

Yeah I hear ya. I could always go pick 'em, but damn I want 'em domesticated!
Besides, consider the upcoming legal market - selling semis by the seashore, that would so rock...

So I've been reading a bunch and it seems like having dedicated containers especially for soaking and coir prep might be a good idea.
Is this overkill for most of you? I don't have a whole lot of room for extra stuff I may not need.
Is it ok to soak grains in a well cleaned pot? As long as I don't use it for coir prep correct?
Thanks in advance!

Yes. I’d personally not use the container I soak grain in for coir prep.. sure, you can clean it but the first time you get a contam in a tub where you used one container for both, you’ll be wondering if that was the vector. I do it for peace of mind. Just get buckets… they nest inside each other so space isn’t really an issue for storage and they’re fairly cheap and last forever and when you’re done using it for myco work, it’s still a bucket that you can bucket things around in

Yeah I get a little paranoid using the same bucket.
I always end up scrubbing it really well and wiping it down with iso before making the coir.
Has anybody ever paid attention to agar nutrition affecting jump off speed? A few weeks ago I knocked up a bunch of jars that just sat around, with one eventually jumping off after two weeks and then growing normally. I figured it was because I had been keeping my plates in a room that never got below 90 and regularly went above 110.
So I took wedges and grew them all out at normal temps. I knocked up a new batch three days ago and nothing. They look like it was three minutes ago. The only thing I can think of is I’ve been using slightly harder and more nutritious plates lately.

I’m new to agar . I know I’ve had a great first run with only one out of 50 getting contaminated and most started pinning or growing in vitro . I made multiple transfer and ended up with a few good plates in the fridge. I used rye soak water undiluted with 1-2% honey and 10 grams agar to 500 ml liquid. I made no pour dishes. I would also be interested what you discover as to the reason for the stall or lag . Is it only cubensis or other fungi also? My lions mane has stalled . It’s the moon and the season. The organisms can sense the seasonal change . All my spring / summer grows have more “ muchness” Could senescence be a factor? Is it possible your mushrooms could be mad at you? I believe the fungus is way more complicated organism then we give it credit for.

Anyone got a rough colonisation time for a quarter of an agar plate into a half pint jar? Thinking about doing this for some G2G. Though I might be better off using the pints I have laying around. Wanna make the most of the quarts and leave them all for the receiving end.

Consolidation depends on a few factors. Genetics, temperature, moisture content , gas exchange and other more subtly nuanced environment factors like temperature differential between upper and lower grow room temperature I would say for me a quarter wedge in a pint jar of rye would be ready to go in a week or two .

It's going to depend on if you drop it as one pice, cut it, or make softer agar and shake it in, also if you shake it at 25-30%. I would shoot for 10 days or less with a 1/4 plate, but I would cut it into pieces and shake it at inoculation and 25-30%.

Yea, dry grains have lower thermal conductivity and will not heat up properly, acting as insulation. This could lead to bacterial endospores surviving.
