I'm still finding swiping swabs to be a poor use even when wet first.
I really think the best way to go is rub them out in water and use the water like OvenBag does or wet them and then pick pieces of cotton from it and lay the cotton on the agar.
I'm getting like 1-2 sad little colonies from a huge zigzag all over the plate - Thick rhyzo mycelium to pick from with laid fibers.

I haven't had trouble yet, but last time I just shoved it in the center which made it kinda hard for the first transfer. We will see, I just slid it on a 1 inch line to the edge from the center a few times. I've made syringes with swabs, they work well, now I'm just too lazy to make a syringe. Made some yeti and a white rabbit f2, got a apeXpf on a t3

if you spawn grains mixed with coir to quart jars, do you just get smaller flushes because of surface area, but more total flushes?

Pretty much.
Ime you’ll get a certain amount per quart of spawn and it’s just a matter of how many flushes it’ll take to hit that number.

i dont really use swabs except for varieties that dont drop spores, but i absolutely love streaking spores with a loop in a serial streak pattern
always get tons of different colonies unless the spores are not very viable

yeah, streaking spores from the gills is the best way to make successive generations. Ill make prints or swabs to save spores from a certain generation but always streak from the gills to move on to the next one

anyone notice 2nd flush and maybe beyond have more fibrous stems? im thinkin the first flush is probably the most tender and the subsequent ones are more woody.
ill probably be keeping the first flushes in the "top shelf" pile.

Hey guys, question about genetics… I noticed someone said a while back that they prefer to take a clone, then when that culture grows out, take the spores, and keep doing that- clone-spores-clone-spores and so on… my question is do people just do that because they’re pheno hunting? If you want to really isolate a big fruit isn’t it better keep taking a clone of a big fruit after growing each culture out- I now know that multiple sectors still appear even from a clone, but if you really want to isolate a big fruit, shouldn’t you take a clone, grab a sector, transfer it to grain or another Petri dish in an agar to agar transfer?

To my understanding people take clones to reproduce the fruit and get more of them in order to get more spores to work with in order to stabilize them. In theory if you had a monster fruit and took its spores and then had the same monster fruit resulting from those spores then you could potentially stabilize those genes after multiple generations.
Most of the time people keep clones because they’ve known to produce a lot of identical mushrooms reliably

If I remember correctly it goes:
Spore Grow Clone Grow Spore.
Repeat.
It’s a little more complicated but yeah. You would want to clone and print only the pheno you’re after.

that's for genetic stabilization. IMO there's no real point in isolating a clone culture. if it gives you the results that you want then all the sectors/strains present are working well together and there's no need to separate them
