I like this! Never heard it before. You could bisect (or whatever ratio) the plate and skewer the pieces on the scalpel for the drop.
Interesting. Thats a good idea. Im colonizing 100mm plates and using #11 blades.
Do you think I can skewer a whole plate? Id definitely try, anything to practice. At the first jar of a 50qt SAB session, I got a really good idea for what I got myself into. It made me nervous and now im just hoping for the best.

Skewering the whole agar puck can be tricky, specially if the puck is big like in petri dishes. The red-hot scalpel will melt away the agar and I dropped lovely agar cultures in the sab because of this.
If the plate you use does not allow you to do the tiger drop like with glad minirounds, the best you can do imo is cut the agar puck in 2 or 4 parts (depending how big it is) and then make sure each wedge gets properly disloged from the plate, then tiger drop.

They don't you'd have to go to an auto shop AutoZone or w.e u have around you
my plan for my 90 mms I got are to cut them in half then lift with the scalpel by going under it

I've been using more in my G2G's and I like the speed.
1 whole plate to a G1 jar
1 G1 master to do 7 G2's. Sometimes I'll drop this to 4 G2's if I have a lot of spawn.
When going from G2 to G3 I use 1/2 of a G2 to (1) G3 2 liter. Makes about 1450-1500ML of finished spawn.
As for yields...14 liter sterilites, unmodified open tray culture...Cased... 1500ml spawn:6 liters CVG=about 4 oz dry on three flushes...or more if more flushes happen.
Pretty happy right now.

Should, depending on the thickness of the medium. I've dropped 4 pieces skewered...I flame up the handle of the scalpel a ways and slide the pieces off using the inside edge of the jar but never feel comfy with this technique. I'm curious to hear what others have had success with when it comes to petris. I use #11 as well.

That's amazing, I wonder if it's something your doing different or just the strain you have.

Best I ever did was almost 4.5oz dry off 2.25qts from my 27qt mini in my sig
Seems like I'm just about on par with that with my tubs. I'm going to start using the micropore dial in method from your thread, so hopefully I'll get better pinsets now.

You don't have to have the red, you can only get it at auto part stores and shit. I was buying oil at the time
You can use any of the clear kind as long as it's 100% Bodhisatta was just talking about that recently.

Make sure you do sand the tops tho! I decided not to listen to that advice and have to re-do 24 lids now cause they popped off in the PC.
For some reason the first lids I did are still running strong, 3 cycles in with no sand. But I guess just because you can get away with something sometimes doesn't mean you can get away with it all the time. Best to just do it right the first time

I have a few lids I didn't sand. they're still alive. They won't last as long though
And As far as inoculating shit with agar, since I use these:

I just stick my scalpel in the middle and the quarter section lifts right out. Less agar, less parafilm (including wrapping), less space, less work blah blah blah... It's pretty efficient.

Here's what I'm working with.

The lid to the right is the lid the SFD originally popped off of. The lid to the left is an old PF lid. I could easily cut that disc in half, if not three pieces 3 straight cuts up and down to cover small holes. I have nails that are slightly bigger than those holes pictured. Plan to heat with torch and melt hole in plastic, then sand.
This all assuming I can get that sillicone off the filter. I've not messed with it. Will it come off?
Thanks!
About to fix shit and really get this boat sailing.

Posting again and adding onto this:
Quote:
Speaking of agar: how bad of plates do you guys try and save? My old APE plate I just made transfers from exploded with white growth like 5x faster than all my other plates. I'm 100% sure it's bacteria. I think I'm seeing green in one. Is it even worth trying to transfer with something sporulating?
I took transfers from plates from the fridge the other night. 3 from an old PE plate, 3 from an old APE plate. All 3 APE exploded in weird growth and all 3 PE have green satellites. The plates that came from other cultures are all doing well with no signs of bacteria.
So, is it even worth taking transfers from these? Am I always going to kick up mold spores into new plates? Seems like I should toss out the old ones for sure.

What about just a couple layers of mp tape without the rtd and filter? Or are you looking for something semi-permanent?

Well, I moved my jars a few days ago off the windowsill to a nicer location and the mycelium is growing at a much more realistic rate now. Who would've thought cubes wouldn't be a fan of being next to a single glazed window in the UK winter?
Also, are 20cc syringes big enough for blenderless LI? What gauge needle is recommended?

Yeah I'd say they are ruined if they look like that. I put silicone on the outer part of the circle
maybe soak it in iso and then try to peel it off.
One more thing, why don't you just cover some of the holes in the pf tek lid with silicone and make one hole bigger and cover it with one sfd?

I'm not trying to use the PF lid. I'm just saying hole size vs what I have to work with, i could turn these 7 discs I have into like 21 plastic lids.
Holy shit. I just was messing with it for the first time. I could've licked that silicone off with my tongue. Man. I bet a hundred bucks they all have shitty seals and no wonder it isn't working. It literally just rolled off the disc.
