Weird thing...I got a mold (trich I think) on a pf tek plate that was nocced with a spore syringe, I didn't bother to open it in the sab to salvage the good myc so I sterilized it for 30min at 15psi to kill it dead.
Well now I've reused that plate for agar and is being kept inside a tote with more colonizing agar plates. I just opened the lid of the tub and that disgusting mold smell gave me a punch in the nose.
It smelled like live mold, but this mold smells particularly disgusting. I've taken that plate out of the tote, the stink was def coming from the same plate.
How is it possible that the smell lingers even after sterilization?

I followed Franks tek for my slantz. He used vacutainers in his tek and said to let the cap jiggle loosely for GE during sterilization, so I screwed on my caps all the way and then just unscrewed them a half turn. First thing I did as soon as I opened the PC was screw the caps on tight. After you transfer myc to them, leave them loose and wrap with parafilm.

At work we have clear pp5 rounds with clear pp5 lids the size of dinner plates. Giant petris dishes in my eyes. I can't stop thinking about a culture the size of a pizza...

Yeah really. You near a KFC? Their mashed potatoe cups are PP5, and they're massive.

Kinda wanna take a trip tomorrow. This is the longest I've went in over 8 years without herb (4-5 days) and my head feels so clear. (Trying to pass a piss test for a "man" job on Monday, make under 200 a week now, have the opportunity to make over 600-700 a week, old lady can stay Home with little man) I've been so much more outgoing and friendly lately. Smoking often makes me such a hermit. My psyche has never felt as great to trip as it does now.
Curious to see how a clear headed trip is.
99.9%of the time I'm stoned. I hate a lot of shit. But my head feels good now. Think I have some shrooms somewhere I can find or i bet the bottom of the dehydrator has some cap pieces.

I wonder if a giant agar plate the size of a pizza would be good for isolating? Let it grow out about a foot and then grab a tiny piece?

Who knows man.
I do have a question though.
I have a 7th gen Ksss plate. From my first ever agar run. I've transferred it 7 times in Id say the last six months.
Is it any quote unquote "better" than a second transfer? Or even a clone plate?
Why would I keep transferring vs not transferring. Clean is clean, right? Am I getting "better" "Clearer" genetics by continual transfers, or no?

Lol I was kind of wondering the applications it could be useful for. Or if it would be impossible to work over something that large unless it was grown close enough to the edge.

No, it's not any "better'. Until it's tested you don't have any idea what any 1 single strain in a mixed culture could do. Isolating just limits the genetic pool further. An untested isolate is just that. It could even refuse to fruit.

well a clone plate would be quite different than a second transfer.
You are definitely getting more specific genetics isolated by the 7th transfer, however that doesn't make it "better". You may have isolated complete junk genetics, all you've been able to select for is how it looks on the agar.
Can I ask if you have been using different agar media on each transfer?
You have a much better shot with a clone. If you are going to be doing 7 transfers on agar, you should be taking more than one transfer at each stage. So that by the time you get down to relatively isolated genetics on the 7th transfer, you have a wide variety of strains with nearly isolated genes. This way you can grow each out and keep the best genetics. The advantage is that because each plate will be already an iso, or close to it, the clone taken from from that grow will more certainly be identical in performance to the grow itself, which is not completely true of cloning from MS. In fact, you wouldn't even need to clone, which may help prevent senescence. It seems to me it would be a lot easier to just clone from limited MS. limited MS meaning you do a MS grow but you a least transfer once or twice so that you aren't throwing in thousands of strains, but rather just a dozen or so.

No, not really. I use PPs and probably wait til it's a silver dollar sized. A fully colonized petri seems like it'd be a pain in the ass to get in the jar without touching the sides. I'd rather make a single circular cut and get it in there (or Tiger drop).

Hmm, I'm sure now got a Spot with 'green' mold in my 2nd Mono bad work, but the pinset was so nice, damn. The first flush is still growing crazy awesome - Just let everything grow ? Toss the cake after harvest-time or trying for another flush? The mold isn't that fast than I am used to, so I think the first flush is safe, but not sure with number 2.

Thanks for the response! Ill probably let them grow out a little more then drop em in a jar of water. Ill cut them up if I gotta. But thats very good to know.
Im relatively new to agar, only a few months of it under my belt.. what are your colonization rates like? I understand I might have isolated clean, slow, rhizo growers. Just curious

Here's an old APE plate I've had sitting around for a while.

1) I know it's agar, but does that look like APE to y'all?
2) Still worth transferring the pin to new agar?
