As long as you slice both ends of a stem with sterile scalpel in sab or flow then push needle down through the middle of the stem it should be clean

I have neither of those genetics at the moment. I am at the gathering stage. I have many kinds of cubes at my disposal, but none that would cross and provide something viably unique enough for me.

I saw the biopsy poke from tub fruit to LC on YouTube (I know I know). It was oyster not cubes
I'll give it a try along with tissue to agar when I can.

I know this is a rare case and most likely an exception to the rule. This is a germination plate from a ess swab that I took myself. The plates that I used the zigzag technique on came out looking really good although still a little chaotic. But the ones where I just dabbed the tip of the swab onto the agar came out like this.

I've come across an old thread here where one user suggested that when a spawn jar is difficult to break up (against a tire or similar I mean), it may be a sign of bacteria. Is that true ?

It can be an indicator that something is wrong... not 100% of the time tho... ive had spawn that was rock solid, once spawned went for a good 3 flushes before i tossed it for a fresh box.

Thanks a lot. I'm asking because one of my jar definitely would not break up no matter how crazy I went at it, and the tub that used that jar has what looks like bacteria.

Its possible then... generally people will let bacterial tubs run... whether they need the fruits or just doing it for science... moldy tubs on the other hand, should be discarded immediately, IMO.

Iβve inoculated with spore plate wedges a bunch of times. Usually when itβs something Iβm excited about so Iβll take t1s and drop some of whatβs left. Works most of the time but you can definitely have contam show up way later.
Thereβs quite a few posts Iβve seen about spore tissue straight to lc or grain too. Iβm not quite that brave yet.
Iβve been thinking about trying a Melmak/ufo cross just because theyβre as close to opposites as you can get.

Should I worry about whole oats having bits of straw and shit in it? I cleaned out what I could when I rinsed but theres still some in there.

Look for wheat too. Where I'm at it's 20 bucks for 100 pounds of wheat, rye is nonexistent.

How moldy is too moldy for you? I'll let the occasional 50/50 split tub fruit till a whole mushroom is consumed by mold. Last tub that was practically 50/50 mold and melmac; I got 6oz dry yield from the clean fruits.

As soon as I see it... lol. That shits going in the garage thats currently about 40*F at the very least... I don't like having any green anywhere in my room... I know if your (other) spawn is clean it shouldn't matter but im more "better safe than sorry" on it.
