I think I see a pin on one of my cakes that I've been moaning about not being able to get water drops to settle on. They've only been in the chamber 2 days.
I'm basically RR and Bod combined.

I had the weirdest thing happen to me. I have 10 jars inoculated with 8 different cultures and not one of them has grown and in fact they all look dead. Its never happened to me before in all these years and wanted to know if its has happened to anyone else?
All the jars b4 and after are fine..... Odd
White millet.

how did you inoculate?
With agar culture wedges. Pretty big sizes too. Its crazy......

Has anyone used a seemingly clean ms germ plate to inoculate grain and had issues later? Or will contams always present themselves on agar first

You should always do at least one transfer to be sure.
A clean germ plate is very unlikely. Competitors can be extremely hard to spot in that chaos.

Agreed. I usually take my scope to them and find good spots to josex poke em.

I did take a T1 to another plate, but that was before I saw any nice rhizos

I usually go to T2 before I'm completely satisfied but that's just me. I've put T1 pokes to LC before when I was in a rush to have fruits to clone though which is probably the dumbest thing I coulda done so you do you.

I have T4s on LME 20% agar that are 100% Tomentose with no signs of rhizo but grow rhizo on coir or brf cakes immediately. I just want a pretty photo on a plate :'(

Man I guess I need to catch up. I remember seeing some people talking about Josex poke a while back but seems like everyone is rocking it was great success these days. I need to see what this is all about. It's just about T-time; need to do some reading. Yay learning!!

You can poke a fruit directly to clone in LC too right? Not having ever fruited my plan when I do is.
Poke biopsy to LC Scalpel to agar Spore print
Still haven't yet established why I'd need all 3 but seems a good way to experiment.

Definitely take some tissue to agar like you said and spore print so you can start over if need be.
Can't say i've ever seen anyone say they went straight to LC from clone tissue... seems like a recipe for disaster.

I think the comment on Josex's poke tek was refering to in vitro fruits. Don't think you would want to do that with a normal mono/shoebox/cake/etc. fruit.
