Sh4dows, I make sure the lid is as level as I can make it and tighten it down before turning on the burner. There is absolutely no leaking from the lid while its running. No. I have never used vaseline or any other lubricant. Tbh, I never had a problem until I tried to stuff 6 bags into it.
Cullingman360, the AA is fine. The only damage was the plastic stuck to the walls. I was able to get about 90% of it off. I'm sure the rest of it will wear off over time.

I had to change out my presto PC pressure gage because of this issue. It wasn't reading right, telling me 13psi, but by all means it was knocking at 15psi obviously. Nothing wrong with my PC at all other than the gage being fucked up.

I have been working with agar since I started with five or six different varieties, in addition to staggering some (relatively) successful MS grows using PF tek, BRF to bulk, and MS to rye shoeboxes that are all in various stages currently.
My agar plates (started from MSS) are a lot more uncertain however... getting into the T-4/5 range they are looking better (and I had one nice T-3 I was able to spawn with).
My question is kind of subjective, but is it worth the effort of continuing to pursue these original agar isolating projects which are getting closer to "clean", or should I just switch entirely to cloning samples that are known to fruit? I am finding the one or two clones I have taken show more vigorous rhizo growth on the plates anyway and will likely require less transfers.
Maybe I'm just putting the cart before the horse...

clones will often show the vigorous growth simply du to the switch from tertiary mycelium back to vegetative. if you have clean plates with limited genetics I would run them and pick clones. ya never know when your missing winners just looking at plates

Boy howdy, just looking at plates. It's a real learning experience. And some white stuff looks like right stuff. And then one transfers and transfers... And on and on... I could really use a success, I'm thinking go back to RW or PESA.. They gave me π...

Hey cron. Is there anything I should do on agar to help dikaryons co-exists? Look for a certain pattern ?
I am trying to do the exact opposite of everyone else and combine a diversity of genetics into one spawn culture.Multi Isolate

Dikariotic myc will cross and share genetic material, anostasmosis i think it's called. Someone fill me in on the right term if you know it please.
Or maybe i misunderstand your question cause you say your not trying to cross, but the post reads like you wanna mix up the genetics and see what you get. You're gonna end up with something like a pundant(sp?) square with different combinations of dominant and recessive traits. You can than work to stabilize the traits(or see more recessive traits expressed) in subsequent generations.

Oh i see. I don't think there's really a way to accomplish that. 2 varieties in the same tub you could definitely do by just spawning different grains on either side of a tub and don't mix them together. But coexisting in a single grain jar and handle being mix up doesn't sound plausible. you might get a few of each pheno, but many would mix genes.
Or maybe i'm about to learn something new here to.

If you can get a couple clonesvthat form a wall on agar without throwing off a third sector they will work in a jar

I recently bought a few tubs and I dident notice they have a small opening where the lid meets the handles ... think I could still run them unmoded? Or should I try covering these holes with saran wrap

Although itβs been shown that you can dedikaryotize the myc via blending in saline solutions. I know itβs been done with Pleurotus before but I think Sidnee Oversinglinton also did it with cubes.
