If metabolites are present on a plate does that equal bacteria or some sort of contam? Or could it just be digestion?

It's highly unlikely that mycelium would be producing metabolites just from digesting agar. I've personally never seen it on a healthy plate.

It's yellow when I look through the bottom at the growth- no yellow anywhere else. Looks like I have more cleaning up to do then. Thanks for the reply

Agar does change color when it's being digested... Sometimes to a yellowish color. Are you sure that's not what you're seeing?

Didn't know the agar could/would do that. It's definitely possible. I am prone to overthinking and have admitted failure at times when I did not even have a problem.
I did dye the agar green btw

Okay, that's a factor for sure. Especially with dyed agar. I'd say if the yellow is under the myc and it's kind of changing the color of the agar, then you're probably fine.
This was dyed agar at one point, either purple or blue:

Yep. Every single one of my plates are yellow before I transfer. Let them plates grow, and that's what happens. Softer agar IME yellows quicker than stiff.

So when we talk about testing cultures what is your go to method. You take a pin clone transfer it a bunch of times, you have 6 clean plates, whats next?

Putting it to grain then spawn.. run like 10 to 20 tubs if you know youre giving optimal conditions and it just keeps pushing out shitty genetics you toss and start over the more to transfer will be just a waste of time you could get better results but its best to just try a new culture

Yeah really I need answers too. Is the 6th transfer of a clone gonna be better than a second transfer? Why or why not? You'd think it's narrowing it down to thinner cleaner tighter genetics with more transfers, but mushrooms are weird as shit so who knows

It can be better or worse.. depending on what you've isolated and what you've isolated out, if anything. I wouldn't assume that you're isolating genetics out from a clone plate unless you're taking really ridiculously small transfers.

Well ya but if shitty genes have been proven on multiple grows those are more then likely the dominant ones ... kinda like black peoole wanting a white baby they could make 1000 kids and its a possibility they get a white baby so they just end up adopting or killing themselves either way would of been better to adopt haha
