I think colonized slants in a dedicated lab fridge (no temperature swings) are probably the best way. You might still have to transfer them to new slants periodically...

Thanks mush.
Pasty plates arent very good for long term storage.. in my experience they dry out after a month or two but they do still transfer fine at that point so you can probably get away with a transfer every 2 months if you want to keep a culture. Slants are quite a bit more ideal for long term.
For long term storage via pasty plates make the plate higher nutrient so it doesnt try to pin on it as well. You can store it on a shelf without needing to do anything or if you wanna try to make it last longer than 2 months you can put it in a ziplock bag and then in the fridge, saranwrap the outside of it if you feel inclined so it doesnt dry out through the filter.
I've been using pasty plates since I started basically and I find them super effective and easy to work with. Hope they treat you good too..

As per RR. STORING GRAINS/PETRIS - Actually, grains have been found better for mycelium storage than agar slants, but unfortunately take way too much room in the refrigerator. However, I've used jars that have been stored fully colonized in the refrigerator for several years. Put them in the refrigerator at full colonization, and then when you remove from the refrigerator, allow three to five days at room temperature before spawning.
I don't bother with slants at all after reading that but there is as RR said the problem of how much space they take. Having said that with RR saying grains are better than slants nothing is stopping you from downsizing to a pint jar of grain. Do it with WBS and everytime you need genetics from it just use sterile tweezers to put a piece of milo on agar lol. That should last you til eternity.

Cool, I've decided to give Spitball's quick tek a shot. Got the rye draining now.
For the jar lids I figured a couple of holes covered in 2-3 layers of micropore tape should be ok for GE?

So...what about holes in the lids? Would it be better to used original lids, or can I get away with a hole and filter disk? Thinking this might be another good idea once I get something nicely isolated. A couple of my MSS jars are growing quite well, and I"m thinking of preserving

With a propane burner dump in rye fill with water about 2-3x the depth of the rye. Turn burner to medium high and set timer for 40-45 minutes. At that time the water should be hot but not boiling. Cut heat and set timer for another 45 min. Rye should be hydrated with no burst grain. Drain and spread out to dry while still hot. Should be ready real quick if u spread it out while its still steaming.
Thats what I do anyways

Okay I don't think this deserves its own thread so I am going to be revisiting spawn bags and trying to unfuck whatever I kept fucking up.
I went and got 100kg of wheat. I have Unicorn 0.5 micron XL bags.
With information from Pasty and company this is the plan and speak now if I'm already fucking up.
Wheat into pot, boil for ~20 minutes hard. Pull a spoonful and check to see if the middles have hydrated. Drain into my fuckit-buckit strainer then immediately dump onto my 4x4' strainer. Let steam until theres little to no moisture on outside of hulls.
Fill bags each with 4 full quarts. Place aluminum foil from bags mouth to grains so the bag does not stick to itself. Tyvek not necessary (Pastytip) No pre-seal. Roll air out of bags and accordion so filter patch stays dry. Place layer of rings in bottom of 23qt Presto. Place trivet on top of rings. Make one layer of 3 bags if they will fit. Accordian folds up. Make second layer of 3 bags if they will fit. Accordian folds down? Place two plates (Mine aren't heavy on their own) on top of bags. Fill cooker with 3 quarts of water as per manual. Wash then lightly lube my gasket ring as per manual. Stove to Hi. Wait for constant jet of steam. Let vent for 10 minutes. Place weight on. Let pressure build to 15psi and fast rock start. Roll back temperature to normal operating and let return to 15psi. Give it three hours. Let cool in PC overnight (Is this necessary? I'd like to do two batches)
Next. Remove from PC, keep accordian folds closed so I assume I can just put the bags upside down on a clean counter. Fire up my flowhood. Showered, facemask on. arms washed with isopropyl, gloves on. Unfold bag infront of flowhood. Let sterile air gently form a plenum and give a spritz of CLEAN LC. Bunch top of bag, zip tie it closed. Move to next bag.
If there's something wrong in these steps speak now or forever hold your Reese pieces because I'm giving bags another shot based on some Shroomery advice from another thread and I'd like to have a simple as a pimple repeatable success here.
