Im about to tic tac toe 6 jars. Should i cut each plate before it's jar or cut them all up then drop them all

6 plates to 6 jars? Cut them individually right before dropping, fewer movements.

Hello Everyone,
not sure if this is the right place for this question. I have a AA 25x and the steam gauge has been reading off since like forever. I still worked with it, but finally got to order a new gauge the other day.
Is the change of the gauge straight forward ? Should I use Teflon thread tape ?
If you have some hints what to look out for would be much appreciated. Or if there is a tutorial somewhere. I plan not to ruin my AA if possible.
While I'm at it a question less important: The steam gauges they use seem rather cheap. Do you guys know of a better steam gauge that is still affordable and fits the thread (I think 1/8" NPT ?? ) and survives the conditions ?

Ran my pressure cooker for the first time since moving from elevation 909’ to elevation 18’. What a treat, knocked off about 30 min of warm up and was near silent compared to the higher elevation.

Got some tyvek. In your guys' experience, what's best to use. rtv or micropore? I can see the rtv lasting longer, but I realistically don't know how long these filters last. I'm just tired of pulling micropore off these lids everytime I'm running agar which is twice a week at this point.

I have some pan spores germinating on agar, but I was wondering how long should I let them grow on the original streaked plate before transferring them? Is there a rule of thumb?

For germination plates, it's normally advised to transfer as soon as is feasible. So as soon as you get some good healthy growth

I like to take transfers as soon as the points of germination "fill in" if that makes sense.
You could say once the white fills in the point of germination. However, I dislike adding the white part, as sometimes I have germination plates that fill in really white really quickly and sometimes my germination plates stay a little more translucent than you would expect on like a T2 plate until they grow out a good bit. If that is the case, waiting until they fill in really white normally means I've waited longer than I'd like to take a transfer.
If that makes sense.
However, if you take a transfer before they've filled in white, make sure you are confident with discerning the difference between a weaker or germinating myc versus bacterial film or some other mold. Its normally a fairly discernable difference, but I made that mistake some while starting out. Keeping my germination plate and looking back at it helped me in that regard.

Yeah makes sense. I just lack the experience to make calls with confidence!
Like I said my camera isn't the best for close up stuff. Or anything these days, I need to replace it.
However, is this sort of at the 'filled in germination point'?

Wicked, excuse my hand holdy questions! I appreciate the help. So I grab a couple of those dots and drop them onto a clean plate like I would when making a transfer from the edge of growth?
I've only worked with clones so far.

use micropore. Easier to replace the tyvek that way, and you don't waste the rtv. Save the rtv for when you get sfd

a magnifying glass is a great tool for looking at small growth. i use a 16x loop.
