140-170 used to be the standard however, people’s specific elevations often throw a wrench into the low end of the temps and 170 is a bit like riding a razors edge. I personally like to keep it between 150-160 for about 60-70 min. 90 min pasteurization is unnecessary and may even contribute to partial sterilization (especially for people living at sea level.
With bucket tek do you remove it from said bucket after 60 or like me just leave till you need it?

Look for a seed supplier. The best and cheapest rye here comes from a seed supplier. Seed grain seems to work better than feed grain imo.

Ever since I built my hood, I've been having trouble with A2G and G2G transfers. I've concluded it's not my hood (thank god), because I've left exposed agar plates all along the surface of my table, and also on the tops of my grain jars for 30 seconds with no contaminates in nearly all cases. When I was working in a SAB, my success rate was consistently 90-100%. I don't have any issue with A2A transfers, but everything involving a jar leads to a little over 50% success.
It's most likely my technique, of course, but I just don't see what I could be doing wrong. I always wipe down my jars with an alcohol soaked towel, flame my scalpel after every transaction, and I transfer at least two additional times after getting a clean plate before doing A2G. I inspect each jar daily and pull anything that looks less than perfect immediately. Generally, for A2G, I just cut a third of the plate and drop it into the jar, until the plate is gone (perhaps this is a vector and I'm sloppy here).
I've been trying to narrow down the causes, assuming it's not my technique, so I've come up with the following list of probable vectors, if you could tell me what you think is most probable or maybe you've thought of something I haven't, please let me know!:
- I cook my grains for 3 hours now, rather than 2-2.5. I'll see how this helps. Previously I was having jars I never opened contam after sitting for about a month. - I use unmodded plastic lids, and my jars sit on a shelf directly behind my hood, so the air from it would hit my jars at an angle perpendicular to them, so I'm thinking that perhaps the hood shoves spores up through the threads of the lid. - The plastic lids often come out of the PC very loose, so contams may get in during this time (but no one else seems to have this issue). - Generally, I drop the entire wedge from the center of the plate all the way to the edge, perhaps I should to excluding the very edge, where contams often reside.
If you can think of any other tests I could perform to narrow down the cause, I'm all for that. Meanwhile, I guess I'll start incorporating a SAB back into my workflow unitl this is resolved.. I'll get a video next time I do some A2G, perhaps even later today. Should I make my own thread for this, or is there a 'technique critique general' I could post in? I'll also include an overview of my lab space so you guys can get an idea of what the room looks like. It's driving me absolutely insane and has been going on for months now.

I need help ASAP. My PC isn’t blowing steam from the valve and I’m wondering if it’s okay to run it still? I’ve never had it not blow out before but when I put the shake weight on it still builds pressure. Please help ASAP. I am running it right now and it’s building pressure. Just not sure why it doesn’t blow out like it used to. It blew out a bit but not a steady stream.

Most likely the vent pipe is clogged. Use a paper clip or something to unplug it.

I did check it and there was nothing in there. I stopped it and checked everything then ran it again with the same issue of no venting.

The seal is brand new pretty much. Nothing was in the way of the valve. There was steam coming from it but it didn’t last long. It was coming from that thing that wiggles before it pops up.

I’m using a stove top... just took it out of the PC and hope it’s all good. I was running a media bottle full of agar.it didn’t steam out the vent... it did reach pressure though and ran the 20min cycle. I just hope it’s all good.

what brand/model of pressure cooker are we talking about Show me a pic if possible.
It souds like...if the pipe is clear, that you are not heating the cooker up like you think you are (burner going bad, losing wattage or some shit) or are using more water than normal taking longer to get to a boil. If it wasnt building pressure then you put the weight on and it did start and the pipe is clear the only logical physically possible explanation is that you just werent hot enough yet lol.
