There won't be any really significant isolation taking place when you transfer, there will be thousands of underlying and intertwined strains present within the sample. It takes many tiny transfers to significantly isolate your culture.

Ok that makes sense so once I take a clone to agar and then the agar to a slant should I try taking quite a big piece of agar to that slant to ensure the most amount of strains?

No, take a small transfer. Even though the transfer seems small from your perspective it's actully quite large when contrasted with the microscopic threads of mycelium. Just don't worry about losing much is all I'm saying, it takes considerable effort to reduce the genetic diversity of a culture.

Just spawned a tub of PE. How do yall go about trying to avoid a flush full of mutants? Some people seem to drop a late casing and others just leave it go. I just mixed the spawn and coir/verm together and leveled it out like I normally would for a cube.

You're gonna wanna case at around 90% I do anyway, Still get a few anyway. Sometimes I case twice.

What i got from some of RR's stuff i was reading was that the longer mycelium is on the agar together, the more genetic information is being shared. Like even if there are different sectors, if that plate is sitting in the fridge for two months the different sectors are exchanging information and influencing each other for good or ill. Does anyone know if that's the case? I dont remember exactly what i read but that was the impression i got

Ill try that. I wanted to do a lay casing or lay mixing, or whatever its called where you mix the sub and all but one spawn jar together, top it with the last spawn jar, then top it with a sub layer.
You use coir to case?

For casing I use jiffy 50/50 it's peat moss and verm, there is discussion of whether pasturizing is necessary or not. Some use straight from bag.

I might late case with coir, just cause I have tons on hand, but Ill check out the 50/50.
I always forget what I want to do with stuff when spawning. Its like every plan goes out the window when I crack those jars open ,

I think a lot depends on what kind of myc you’ve got.
Most of my subs look pretty rough after the first flush unless it was a smaller pinset.

Reshi block i made a couple months ago Didn't seem to be doing much so i set it outside under our apartment car port Started to fruit so i cut the bag up to let more air exchange My question is, can i leave this on my grandparents covered porch as is? Will they need to "water" the block at all?
*grain spawn to lippa tek hwfp. Attempted to case with coir...for idk what reason

Thanks for the answers, i think i maybe get it out of the tub for the second flush.
I made some LC a weeks ago and test it on 4 jars with a bit of grain trying a grain agar tek, but i had to pour it because the needle couldnt suck it, so i just poured it... 6 days after all jars show micelium and apparently no contams, will try with biggers jars now.
Can i fruit this little of grain? maybe with a mini coir casing? to take a clone maybe or just for fun.

Ive thought a lot about doing something along the same line, or at least the same vein of thought.
Ive been thinking about letting a tub colonize fully, then cut it in half long ways and then stand them up on the cut edge. It would more than double surface area. Also been thinking of other weird ways of doin it.
Wonder if anyone ever tried something like that. Im sure they have.
