But I am wondering if the plate going through this cycle of pinning and dropping spores is the is dish clean until a point where it uses all the nutrients in the agar?

After years of running rookiemode wbs grows interspersed with the occasional pet store sorghum grow (I really loved the performance of sorghum btw) I have finally found a grain supplier that sells to the public. I'm gonna get some sorghum, but there are other grains to try and I wanna try one out.
I am curious as to what you guys prefer: Oats, or Rye.

I've been watching those comments and I'm of the opinion that Wheat is the fav... Only 45 lbs to go on my race oats and I can try wheat!

I like oats, but to be fair I don't have much experience with other grains. Just using what's easiest to get for me.

Inoculation Loops
Best reply: MLPismyOPSEC said: NFI how many microliters of piss it can hold though, sorry bro!

Inthepit said: So this is my loop... Ya pretty ghetto! I hate these wire loops and they didn't come with a holder, fer cryin out loud! And the wire is too flimsy, it just bends at the slightest touch.
So, I searched the Shroomery, no joy...
So, this POS loop is .010 wire diameter, 30 AWG, and loop is .160 ID or 4 mm Mostly I see 24 and 25 AWG offered.
μL, what the hell is that μL? mi·cro·li·ter /ˈmīkrōlēdər/ one millionth of a liter. loop Volume One μL loopful of urine!
Loop diameters: 3mm =.118, 4mm=.157, 5mm=.196 1 μL to 10 μL volume of piss!
In 1960 O'Sullivan published a study carried out in Birmingham comparing the colony counts obtained with the pour-plate method with those obtained by culture of a "standard loopful" of urine. For the inoculating loop method, a platinum wire loop , with 3 mm internal diameter, was used to sample the urine specimen, and the loopful spread over a nutrient agar plate. more article... The standard loops are made from twisted Nichrome 5 wire. Nichrome 5 is fast cooling and exceptionally durable, and the twisted shank does not vibrate as much as single wires.
I'm not a heavy user of the loop, in fact I haven't even put a new one in. But on the other hand I'd like to try different wire gauges. A 4 mm loop seems fine.
So what do you use? 1. A loop holder with fixed loop? 2. A loop holder with replaceable loops? 2.1 Yer own ghetto wooden dowel with the loop stuck in the end? 3. 25 AWG, 26, 30? 4. 3mm dia., 4, 5, loop? 5. A pretty pink loop holder cause yer such a girly girl! 6. An Exacto holder with Kenthal Vape Wire 7. Stripped TV cable wire even though you own a $500 AA PC! LOL 8. Just hold the loop with yer fingers!

Good information from people trying to help, hilarity, accusations. It has it all and more and I give it to you for the outrageously low price of $19.95 and a spore print. Surprise a brotha.

When u guys use the media bottles with the gl45 caps do u drill a hole and stuff it with polyfill or just leave it slightly loose during PC I've seen both but I'm wondering what would be considered better

I loosen my cap and still cover with foil. I still only fill my 500 to 300 though, I've had overflow before.

Slightly loose cap (I do like 1/4 of the twist after it has closed(not too tightly) followed by tight foil. Basically I followed this c10's agar TEK almost to the letter the first time I tried agar and it worked out great. As soon as I open pressure cooker I tighten the cap and let it cool down (either for pouring right away or storage in the fridge). You don't want any contaminants dragged in via gaps around the cap when the temperature/pressure fluctuates during cooldown.
With regards to media bottles - the good ones have markings for the maximum suggested level of fill. E.g. 1L bottle has the line at 800ml which seems to work fine. However, different agar recipes(agar proportion and whatever nutrients are added) can affect how much/quickly agar can boil over but should never go above ~4/5ths.

Pretty much same.
Fill to 400-450ml then slightly loosen the cap and put foil on.

Oh Roger! What a "great" find. I mean for being house bound for so long, this is one hilarious break. The Meaculpa loop Oh man, I just saw a video and he's actually using that loop.
