I keep getting satellite contams in my plates. The weird thing is in every plate that it shows up, it's the same. It's just one single satellite colony roughly an inch or so away from where I placed the transfer in the center. Is it fair to say this is likely due to not properly flaming the blade or handle of my scalpel? Cultures look clean to me. Only the satellite colonies giving me issues.
Here's an example 1st transfer plate, but it's showing up almost exactly the same in my 2nd transfer plates. All satellite contams are roughly the same distance from the center culture on all contamed plates.

Sounds like scalpel or gloves. Do you use FH or SAB? Do to tap?

Yeah I'm thinking scalpel and/or gloves. Just coming back from a long break, so I'm still getting my sterile technique back. That's probably part of the issue. I use a SAB. It's weird cause I've never had such a consistent contam issue. That's how I know it's more than likely some systemic mistake with my technique.

No you're good my man. And actually recently all my transfers have been coming off easily so I haven't had to do anything other than touch it to the transfer plate. I'm about to take some more transfers, so I'm going to be extra cautious this time and see if these come out a little better. I just got a new #7 scalpel handle yesterday that will be easier to flame sterilize so maybe that will alleviate some of the issues. I'd imagine part of the issue is my break from doing any sterile work for around 5 months.

Try wiping the handle down with iso before you start too if you don’t already.

Thanks for the reminder. For some reason I've forgotten to do that since I've gotten back. That may help a little

Can you grow sclerotia from a growkit? I got a growkit placed it in a bag of brown rice that i desinfected in the oven in a foil package. Watered is twice second time with dextrose water. Rice is turning mushy maybe 3-4 weeks passed. I took a coffe filter sprayed it and the bags outside with acetone taped the filter on the bag and heated a needle with a lighter i poked some holes in the bag with. I read that when i got the kit there was a blowoff period when its exposed to oxygene. So i thought ok il take away most of the oxygene when its placed in the bag and let it grow. First week it was giving of some small amounts of white fluid thats turning the rice to mush. Smells wery sweet shroomy ,held the bag upside down excess fluid run trough the holes of the filter cloging it up turning it white and mushroom like so poked new holes. No idea whats going on in the bag. Dont know if you can grow truffles from a kit or how the blow off period works. But i like the smell its wery sweet mushroom smell from the filter. And no mold or infections. Cant tell if anything is happenning yet. There are some white spots in the bag but i cant tell if its the mushrooms or the brown rice that turned to mush. I could not find any info online on growing truffles from a kit so thought il just test and see. Il try to upload a pic later on.

what kind of grow kit? "mush" doesnt sound good. if the grow kit came with a sclerotia forming species then I would assume so. was the bag of brown rice part of your kit? never heard of a "blowoff period" maybe upload some pics

Sounds like a homemade growkits. What ever you have it sounds like yeast and bacteria. I would get rid of it asap.

Are these plates ok?
Quote:
Nigel Stephens said: Hi there, first time with agar, would appreciate your thoughts on these two plates!
These are both transfer 2 from a clone. The clone was originally grown out on MEA 2%. T1 was to the same medium. These two plates are transferred from the same sector. Plate 2 is MEA 2% again - the myc is very translucent but seems healthy to my untrained eye.
Plate 1 is new formulation agar; potato malt extract 2%. It is quite nutrient rich would that explain the tomentose form? I think it looks clean but could be wildly missing something.
Do you think they’re ready to go to grains? TIA

They are. Ok clear thanks, will keep transferring number 1.
For what it’s worth, both these transfers came from something that looks like dish 2 does now, I.e uniform and translucent. Wasn’t sure whether the medium made a significant impact on the form of the mycelium.
What should I be looking for to indicate the myc is ready for grain?

The growkit was from cosmic kitchen in Netherlands. I dont think its yeast becouse the smell was from day 1 when i placed the kit in the rice. When i say mushy i mean the rice is breaking up into a mass.
Uploaded the pick. Cant see whats going on in the middle of the bag where the truffles is. Also the rice brown colour is dissapearing and it turned more white.
