Sorry for the poor quality picture and terrible petri labeling, but does anyone know why a GT print and a Burma print would produce such different growth when inoculated on the same date with the same batch of plates? I did 5 of each Burma and GT and all of the GT kicks ass compared to the Burma in terms of looks.

Well... Right off the bat you started by saying they were from two different prints of two different varieties. So, you have genetics, and different potential levels of contamination as variables at the very least.

Fair enough, thanks. It just struck me as kind of odd how different they are, but then again I'm very new to this.

If it's not contamination causes differences it's usually genetics, especially on agar where conditions play no role.

Hello, forgive me if this has been asked before, but I was wondering how long pasteurized jiffy mix (50/50+) casing can be kept before "going bad". Can I pasteurize a bucketful and use it over the course of a few weeks?

Oh okay. In that case, how do you guys like to pasteurize your casing layers? I typically use the muda bucket tek for pasteurizing substrate, but I do not think it would be suitable for (just a few quarts of) casing.

Hey y'all, first post in a long time but I've returned and this thread is sweet. Anyway, been going over shit and so much has been deleted, it's ridiculous! OP pics and multiple posts throughout the thread are deleted by users, admin, "a la Obscurity".. point being, I've been UTFSE and I'm sad. What got people so spooked? Also, seems like shit's changed over the years, I used to use Doc's WBS and Damien5050's coir/verm in a pretty sweet fridge setup. Looks like everyone just does monotubs these days, what teks are you guys using?

Question!
Has anyone got a link or picture and/or know, How P. Subaeruginosa does on agar PD's?
I have been looking into making/buying a Nutrient based agar plate.. and more making a mix of that n saw dust... I know there will be very high chances of contams, Which is why I want to try and mix the wood/ agar for immunity to that. The Wild patches seem to be able to EAT/(Grow-over) other fungi, moulds & algae..(Speculations from Observing)
Any idea if these could work better than general Agar..? Fungal media Dermatophyte test medium · Potato dextrose agar · Sabouraud agar or Nutrient agar? PD's Nutrient agar is usually used for growth of nonfastidious organisms and observation of pigment production. It is safe to use in school science laboratories because it does not selectively grow pathogenic bacteria. ---------------------------------- Wanna experiment with this... or should I just stick to wood rot/chip & cardboard..? ^^which I want to make a glucose blend of.. ¼ Glucose, ¼ shredded/1mm² Cardboard & the other ½ Wood decay mixes for Nutrients.. Which I'm testing what wood the Myc like best..(inner base Nutrients)
Still reading up a lot that's already on here that I can find.. Just not much with... what I have in mind..
Thoughts? -------------------Edit--->
Google & Ebay FTW... But still would like to know Thoughts or personal accounts.
Nutrient agar From Wikipedia, the free encyclopedia Nutrient agar is a general purpose medium supporting growth of a wide range of non-fastidious organisms. It typically contains (mass/volume):[1] 0.5% Peptone - this provides organic nitrogen 0.3% beef extract/yeast extract - the water-soluble content of these contribute vitamins, carbohydrates, nitrogen, and salts 1.5% agar - this gives the mixture solidity 0.5% Sodium Chloride - this gives the mixture proportions similar to those found in the cytoplasm of most organisms distilled water - water serves as a transport medium for the agar's various substances pH adjusted to neutral (6.8) at 25 °C.
These ingredients are combined and boiled for approximately one minute to ensure they are mixed and to sterilize them. Then they are cooled to around 50 °C (122 °F) and poured into Petri dishes which are covered immediately. Once the dishes hold solidified agar, they are stored upside down and are often refrigerated until used. Inoculation takes place on warm dishes rather than cool ones: if refrigerated for storage, the dishes must be rewarmed to room temperature prior to inoculation. ----------------But will it work out with Cold lovers?

Mushierage said: You could always use the oven. I wouldn't suggest this though for anything other than this type of casing or vermiculite. Don't try it with peat. Also, it takes longer, and requires a higher temperature.
Space was right, coir is great in the oven. The coir tek I wrote myself is about prepping coir in the oven. Coir literally does not give a fuck.

Nothing got anybody spooked, the site's upload limit is terrible and people feel like they have to delete pics to make room for new ones. Though, when they delete pics for teks to do so, I feel that's a terribly misplaced effort.
