Quick question: Do spores from Isolate fruit contain as much genetic diversity as spores from a MS fruit?

Now that is an interesting question.
Iβd guess not but I have no idea.

Most varieties are pretty inbred and stable so for the most part yes. Early lineages will have more diversity though so let's say an f3 isolate of rustywhyte, this spore print will have a bit more diversity than an f7 but less diversity then an f3 from ms

I would think that the diversity, that is number of possible genetic outcomes, would be about the same. The difference would be that you have narrowed it's genetic starting point. With an MSS, you have no idea the potency, size, growth rate, etc of the "mother" fruit were as a isolate fruit would be guaranteed to have at least some spores with similar genetics as the mother, but no guarantee that specific spores you germinate will be the same.
I could be very wrong though

Right. But the tendency to form blobs on the first flush (like the myc has to remember how to make proper fruits) must be genetic, because I've had PE that didn't blob and then I've had what I've got now, PE that sets pins many of which blob, even when cased, and then fruits on the 2nd flush. Rather not have blobs.

Iβm soooo fucking tired of plucking blobs. The potency is great, but I miss the aesthetics of a nice canopy. 6 more tubs left, then Iβm done with it.

Fields of blobs make me want to go around hurting old people for no reason. I fucking hate nice full blob sets!
P.S. i would never intentionally harm an old person (without reason).

I have been having some weird contams lately on jars I thought were healthy but I noticed this today

do you think that grey wispy spot is just thin mycelium, or do I have hidden mold showing itself? some other spots like that ended up growing in thicker and looking whiter but IDK. maybe I'm just going paranoid.

If you blow the photo up (zoom in fully) you can see pretty well the difference, the strands of mycelium on the greyer spot are really, unorganized

Also this... casing it may be worth trying if you haven't already..Frank's got a good tek on it...
I was about to scrap all my PE jars and then I got a full canopy and insane BE from MS and the only thing I changed was casing it.
Almost gave up on the variety for good

I tried everything - coir casing, verm casing, late casing, no casing, thick casing, thin casing - to no avail. Well, to be fair Iβm still waiting on 3 tubs that were cased super thick (1in) and 3 cased with pure verm. Iβm guessing I got cruddy genetics?

Got too many project going atm but I finally got both PEβs and ai want to cross them together. Gonna be tricky to be sure which genetics made it through without doing single spore or serial dilution which is gonna be extra tricky with swabs. Sure theres someone more capable than me out there but Iβll get to it eventually if noone else does. Also thought some ess X MM might be cool to mix up the gene pool a bit.

You try peat? I cased mine with 5050 with a little extra verm and they did awesome. I could totally see writing off PE if you didnβt know it needed casing.
Itβs crazy that a tiny layer of peat moss is the difference between these two. (Tried late casing the blob one for the next flush).
  Same spawn same age.

Let's talk SAB for a moment..... Was thinking about me SAB procedure recently. Haven't been able to get up to it much lately so it's been on my mind a lot. My SAB in a still air room. The table is as far as possible, about 20ft, and no one enters the room expect me to do lab work. My thought is, my SAB is as safe when I first enter the room as is is while I am using it. There are really no changes in air current or environment that would compromise it. I know a lot of you have said you no longer mist your SAB. My thought is, with this set up I feel I can just walk in, do work, and walk out. No need for misting and settling and such. Thoughts?

When you load the SAB you gotta let the air settle again.

I actually keep my plates in SAB after I pour them and I until use. Part of what gave me these thought. Unwrapped plates have sit in there for well over a week (last 2 I used were in there since 12\20) with no contams whatsoever. So the only things I'm loading (lately anyway) are prints, syringes or swabs which can be put in with no more movement than placing my hand inside. Loading jars would need more time but are usually loaded one by one. Someone related note, anyone had issues with spray bottle of h2o developing mildewing smell from sitting too long? That was also part of what started this thinking. Would hate to think I am spraying my SAB with a mildew solution everytime I do work.
Also, good to hear from you, Quirk. Haven't seen you in a while.

SAB just came up in agar envy thread too. somewhere in the past month (since beginning end of Oct lol) i shifted from spraying iso to a heavy iso soaked wipe downs. i spray gloved hands with iso tho, bottle hangs just right on the cabinet door the SAB sits on. went dry SAB early.
mildew smell is certainly bacteria and mold heavy, not a good idea to spritz mush enemy#1 around even fresh made soapy water never caught on with me, i was iso spraying heavy till residue and insta-fogging plates, drying parafilm, etc was noticed second SAB session.
SAB on dedicated table, in corner, never moves and gets covered when not in use. did have this as a still air room as well, but for all mush cult, including the grow- which i learned late and hard way FAE is really required (so recently fixed this blunder and tossed some B+ shoeboxes that couldnt recover.. also using daily made 150ppm bleach for misting for now, things have improved hundredfold.
if moving too much in room, or dog comes in, then will sit in front of it for 15min before removing arm space cover as well if add/remove anything from inside. i'll load it, put cover back, and sit there getting everything else in order for another 5-10
its also why i went huge SAB, 200q, to preload everything. it can easy hold a few stacks petris and a dozen quarts and a work/pour space, with room to spare and less wide shoulders concern knocking the 80q i tried first.
