Second PE tub. 320 just like the first one even though it started out looking shifty.

Definitely wasn’t expecting my best two flushes to be PE.

I realize MS>G is not the preferred route and dont expect perfect/multiple flushes. I have some agar plates in the works as well.
theyre all bacterial, and unless you have actually clean grain spawn to work with on the side of this project, i wouldnt spawn those. Top fruit them. Spawning them to a tub is more than likely going to bring mold into your future work area and put you at a disadvantage.

I don’t know what to look for on oats I’ve only run wbs, but IME a lot of people over hype the risk of bacteria. Unless it’s super bad like those last jars or you have a bunch of clean spawn on deck I’d always give them a chance. Worst case you get one flush and toss it, and I’ve had a couple slightly bacterial jars put out near canopy first flushes.
Just keep an eye on the entire tub, bottom too, so you don’t let trich blow your spot up.

Amazon shipped my scalpels to the wrong state. I was in a hurry so I ordered more. First order just showed up so I now have 200 sterile blades. Up to this point, I have gone through a total of 2 blades. I feel like a spoiled kid now.

2 blades?? Maybe I’m doing something wrong or got shitty blades but I’ve noticed after a handful of transfers and torchings that the blade dulls enough to start pulling myc off the agar instead of making a clean cut.

That did start to happen. But the damage was quite localized on the blade so if I noticed it happening, I just angled the blade differently. That and it really didn't seem to affect the transfers. Myc is myc, it all grew regardless.
Correction: I was currently on my third blade so I guess 3, not 2. Have used probably 60-80 petris and maybe a dozen or so pastyplate-esque no pours. So roughly 30 plates per blade...ish.

That’s crazy efficient.
It makes sense though I guess you could just only use the tip of the blade until it dulled out then start using the next portion of the blade. I’m sure the thickness/hardness of the agar makes a big difference too.

A wedge with no visible myc left on it will still work and recover the same. I might change blades every 100 or so transfers. Sometimes sooner if I'm feeling like starting fresh. Someone gifted me a 100 pack of #11 blades ive had the box for about 4 years and still haven't burned through half of them lol.

Yeah, a box of a hundred blades plus the handle is like $8 on Amazon. I put off ordering any until I got a little deeper into this, but this point 8 cents per blade is cheap enough that cost is almost non-existent, but even if I did one blade per plate, I would still run out of plates before blades at this point

Lol i just recently got into agar and discovered the empty looking transfer thing.
I had a couple plates where I cut weird and either got a tiny transfer piece or what looked like a cut of agar from underneath with no myc on it so I added another one just Incase and then they all took off fine.
For some reason I just hate using dull blades maybe that’s one of those things that goes away with experience. Kinda like how I’m just now starting to throw out good T1 plates instead of saving them since I have piles of things going on now.

That's a hard habit to break. I'm not as bad as I used to be but I can be a plate packrat. Using no pours helped me to break the habit when you only have so many plates and have to reuse you learn to throw out old ones real fast lol. I recently started pouring petris again just poured 40 plates yesterday first time in years. I figure I can find a happy medium between using no pours and petris to cut down on my waste while still having visibility to know what's going on in the culture.

That's actually the reason why I don't use no pours. I can't bring myself to throw out plates, on the off chance that I'll lose everything and this one ify looking plate might be my one chance to bring it back. With no pours, they took up so much damn room after only two sessions of transfers, I was running out of places to put them. at least with Petri dishes they stacked neatly and don't take up much room.

I get hundreds, even thousands of uses out of a single blade. I usually just forget to order more by the time i do teansfers and go a very long time with the same blade.
Anyway feel free to donate me some blades

It has taken me four hours, and 6 attempts, to fold up 9 spore prints and get them in envelopes, address, and stamp. This is the most mobile I have been in 3 weeks. I still can't stand up straight, or stand for very long, but I cannot tell you how excited I am at the thought of making it up to my still air room to do some transfers.

What does a blade look like after a thousand transfers?? I’m imagining basically a charred lump on a scalpel handle.
Also do you guys wipe the blade between transfers? I just started doing that to get the agar off before torching it after a couple chunks of agar going off like tiny popcorn.
