Thanks TedsDead It is actually part 1 of 2 that I never finished. Was going to put wheels and a RFID sensor on the bowl, and a food dispenser that I could drive the Walker under and have it auto fill the bowl. I should get back on that. If I pushed it farther with a raspberry pi and linked it into home networking I could probably make it automatically drive itself to the dispenser every day at a certain time. I will likely not go that far though.

We were discussing it not to long ago caps not many using it but the ones that are like it. I was planning on switching to it. Still am but have a full ass load of oats to finish burning through first.

About to get several things going after about a year and a half, exciting times! Got a colonized plate from a friend chillin' in the fridge and plates on the way to get spores down for CBK's thing, hopefully they get here in time.
The last few times I poured plates I just cooked my agar in a pyrex measuring cup covered in foil, didn't have any problem. I can see why a different form factor would be good with more volume though, I've never prepped more than 20 at a time.
When inoculating plates with swabs, do you generally prepare a separate swap and... transfer spores from one to the other? Or just take your spore swab and touch the agar?
Not 100% on the cleanliness of the gifted plate (nothing looks off, just you know, not mine so who knows, right?) but tempted to put it straight to grain, especially since I could do that before the plates get here.

You could drop it directly to grain now, but then, if something goes south and it doesn't fruit, you have no way to propagate: I take a transfer AND use for grain... if you only have one prepared swab (like a gift), I'd swipe one plate, roll the swab 180° and swipe a second... if you're working from a print, and have sterile swabs at your disposal, new swab per plate... can wrap entire q tips in foil to sterilize, and effectively get 2 inoculations per swab, leaving one side covered in foil until after the first plate, to avoid contamination

I hadn't thought about sterilizing q tips. I am not a fan of my shitty homemade inoc loop. Will have to give this a shot. Thanks for the tip! Curious if you have had any issues with bits of cotton from swab coming off on plate and causing issues later on. I assume of sterilized it should t matter, just a thought that came to me.

I've been trying to track down local Millet for a while now but yeah I think if that were the case I would change grains as well.

Little pieces of cotton that are sterile and covered in spores are not a problem at all. Often, if i want to save a swab for later, I'll just pull a piece off with my scalpel, streak it across the plate and then intentionally drop it off in there.
Contrary to popular opinion, contams are not magical. They don't just appear out of nowhere.

I do that too. I have not done the tests needed to determine whether or not streaking, refoiling and storing is a safe and repeatable practice. So, if i have only one swab of something that i want to be SURE i get going, I play it safe.

Here's a little bit of a confession. I have never, ever, ever had success streaking a swab. I will streak 2 plates then cut off the end of the swab and leave it in a 3rd. 100% of the time I only get growth where I left the swab tip on the plate. Is it me or my agar maybe? I use 10g agar 10g lme to 500ml water.

Your agar is probably too stiff. Dont know about you but I use telephone and if I use 10g/500ml it comes out way too stiff. You should use a softer agar for spore germ anyway

Yea I use telephone also. It works great for spore germ when I'm not using a swab though. I tried it once using soft agar and I couldn't even drag the swab on the surface it just dug in and got covered in agar. Then I had to cut the tip off the swab anyways. That was with 8g agar to 500ml water. I'm not denying that my agar is probably too stiff for spore germ but also I think I just suck at swabs.

If you use spore solution the water helps germ the spores. With a swab you dont have that extra water to help. With a loop, you cool it in the agar which liquifies the agar on the loop which you then swipe to a plate so the spores have that starting moisture.
With a swab I very lightly swipe it in a zig zag patter across my agar. You dont have to apply any downward pressure, just set the swab on the agar and sweep.
