LtLurker said: i don't know how better to describe it and i can't really do the pic edit thing some do. There's like a big water spot taking most of the plate that you can clearly see when held up to light. If you look close at the right angle it probably won't look like normal smooth agar, it'll be "choppy".
Is part of it this darkened, water stained looking section of myc that I have outlined in red here?
Or is it more of a whole plate deal, even the portions outside the myc? Sorry for being such a pain in the ass, but when I look at it in person I just can't see the indicators for contam, even though I know its contaminated lol. I really appreciate the advice thus far though Lurker. Anyone else chime in if you think you can help myesee what I should be seeing

from the pic, it looks safer up top
looks like a film all under the culture and a lot of it around the bottom
like Lurker said, how does the myc look viewed from the side/angle? is it laying nice and flat or choppy like little waves like he asked?

I wasn't able to get it to an angle where I could see the choppiness of the agar, but that doesn't mean it's not there. I went ahead and did a hot pour on top of some of the plates that looked like that. I'm still not able to clearly see the film, other than the agar looks a little rough/not as transparent in some places. Not sure if that's what you all are talking about. I think its more just having newer eyes to spotting contams, not your all's descriptions. I'll have to keep learning to spot it. Thanks for all the help/advice!

I have yet to try the hot pour, how did the process go? just literally pouring liquid agar over a plate?

Yeah it was pretty much that. I let it get down to the temperature I normally pour plates at (~120F), but I read that others sometimes like for it to be a bit hotter than pour temperature. I think I may have poured it too thick, because I poured it on top of each culture until it covered the whole culture/plate. Since the culture is slightly raised above the surface of the agar, it required a fairly thick layer to cover the whole culture completely. Not sure if I'm supposed to cover the whole culture, or just parts of it.
The hot pour layer was roughly as thick as I pour a normal plate, but on top of an already poured plate and culture. There was still a decent amount of clearance space between the top of the second layer and the top of the petri. We'll see how it does

When I hot pour I cover the whole plate so there's a brand new surface, contam free hopefully. If I don't have clearance or don't want to hit the top I'll pour enough so when I tilt it it at least covers the surface of the old agar. This way is not super thick but still provides a barrier to grow through.
And I like it pretty hot.

I'll have to try tilting it next time if I have to do it again. Do you find hot pour works decently well on bacterial dishes for you? Also, when you transfer the new myc away from the contam free surface, do you actually cut into the agar like a transfer, or just scrape some off the top?
Also, I'll try it a little hotter next time

(This is the nerdiest thing I'll say today). Yes, a guy in my mycology club does. It seems to work for him.

TBH bacterial contams I just transfer away from, they aren't fast movers like something generating spores. But yes it stops them in their tracks. Mold contams are the problem though bc surviving mold Mycelium can burrow through the agar. Bc of this I try to just scrape tissue off the surface. I try to pour smooth so that any contams spores will get covered by agar without becoming air borne. Also why I try to coat as much of the old surface as I can including the walls. This is just how I do it there may be better ways.
