So contams get past the saran wrapping? Or you talking bout condensate?..cuz that shit is a temperature issue.

They're separate statements. Your technique is almost certainly to blame for the contams, so get better at pouring, wrapping, and storing. And Saran is not bulletproof (neither is Parafilm) so it's good to add an extra layer of protection. I fucking hate making plates so I'm going to do everything I can do make a huge batch at once and ensure they're all stored properly until I need them.

Working in gloves sux hot shit on a dick. Lysol wiped the hairy balls outta my arms, scalpel, and gloves everytime i pulled my arms outta the sab. Jst like i useta. Wrapped the plates immediatly after transfer (4 at a time). See what happns.
I dig this proper scalpel though. Like a thousand times easier to get a transfer off the blade. Really had no issues. Easier for me to handle as well.
I have a difficult time believing that much of anything is gonna get thru a double wrap of saran. I suppose that in the process of handling the plates, if you squeezed one and released it that may pull contam laden air in, but meh.

It really sucks right now, in hot ass house with no AC & no fan running due to needing the air to be still... hands get all sweaty in the gloves

Shit does get through occasionally. I make 50 plates at a time latley. And they sit around for a couple months by the time I get to having to make more. Occasionally I'll come across a.couple plates that somehow something managed to get in. Usually mold spores. Sometimes it's bacteria.
I also just leave my blanks on top of my flowhood in a stack. The current the blower makes is probably pulling air towards and around my blanks.

Yep. My Pasty Plates will sometimes get mold if I leave them long enough. They don't get wrapped, but they have MP over tiny hole, paper towel on top, foil still on, and in a shoebox tote on a high shelf. That's some Fort Knox shit. But to each their own, for sure. It's not a biggie to put in a new Ziploc so I always did it.

If you dont like pouting plates what situation merits the use of pasty plates over a petri as its seems like more work to prep pastys than pour petris to store in a sleeve. especially if you do all that papertowel/foil work.
How do u get the agar in the pasty plate then? Lol just givin u shit

That's because you do it in a SAB. Pouring dishes in a SAB makes me want to slap grandmas, hard. In the flow I just kick it in my comfy pants, drink a beer, listen to some project 86 or pink floyd and pour some dishes. I rather enjoy it actually. Poured 50 last night.

Hilarious!!! I was just saying how I sometimes leave the donor plate lid off for 5 minutes while I go and get a beer knowing it will be fine. Love agar work now. Before it used to stress me out being stuck in that box.

oh yeah, forgot bout that. So after all the storage precautions you still get contams? And your sure that nothing survives the PC cycle?

I was under the impression that shit cud hang out on a plate for quite some time without germinating.

Fan filter unit, its a laminar flow filter and fan apparatus that is usually put into the ceiling of clean rooms. For our use, they are put on their side and a wooden frame is built around it to convert it into something that functions nearly the same as a proper flow hood, and theyre usually cheaper. Idk, id much rather build a flow hood designed to my needs, but if it's cheaper and jusst as efficient to buy an ffu and mod it, then i might do that. I'm not very mechanically inclined lol
