quick question about cloning.

when cloning a particular mushroom, from a genetic selection point of view, is there any value in taking multiple samples then fruiting and testing them all? or is it reasonably sure that each selection from a particular mushroom is genetically identical? (or close enough as not to matter?)

Thsts a good question. I'm  not sure. But I can tell ya this. Usually when I make a clone. I almost always take tissue from both sides of the torn in half stipe. Just because. but I haven't really noticed any differences. I always label them as the same culture and fruit them as the same.

But now that I'm thinking of it. Sometimes I clone whole fruits. Ans take whatever growth looks the best

For LC's, is it better to leave them alone until colonized or swirl them a lot?

The bags I use are 0.5 micron filter patch bags from unicorn. Theyre factory seconds so maybe thats the issue but idk I just account for the loss of about 1/6 at this point.

@ van I’ve used double cooked grains side by side with ones that only had one cycle done and had success but it was slower for sure, 3 wks vs 2 with the same clone.

i've cloned pins from agar plates. is cloning whole fruit from a substrate a thing? i noticed once, a fruit i picked to clone from a mexicana tray fuzzed up a little bit after a few hours on the table. printed the cap, and cloned from the stipe. i guess i could have put the whole stipe on agar.

You'd probably have a lot of contam to isolate away from. The whole reason for cloning from inner tissue is to avoid the outer (inevitable) contaminates.

Oh u actually double cooked I missed that part. Yeah Def a slower colonization on longer cooked grains for sure.

I thought u just meant your grains were kinda dark.

for sure it's likely but how bad could it be? the mycelium is already running. there's presumably no active mold. bacteria everywhere, but i notice bacteria tends to stay put unless there's water on the agar surface. so if placed carefully, mold spores wouldn't have a chance to germinate before it grew out enough to make some transfers away from the bacteria which would be easy to see.

i've had some luck taking clone tissue from inside, but it's a pain in the arse with the way the stipes grow twisted. i don't know, but i think i'll try this a time or two and see what happens. you know, for science.

People often repeat that the top of the fruit has less active strains, I believe, but that might be repeating RR's shit. Regardless, taking tissue from the bottom of a cluster is what many recommend because it's meatier and easier to pull from.  AFAIK, nobody has proven the strains that around here.

IIRC, you get away with dropping pins because it's smaller (less surface area for bacteria/nasties) and younger, so faster and will outrun stuff. I've dropped small in-vitro fruits. I'd be afraid of contams that aren't immediately visible sticking with the fruit/myc. Seems like you could just tear apart the outside of it like a banana and then drop tissue that way, no?

One thing I'd mention is that you only need a tiny sliver of tissue , like a scrape, and not always a chunk cut out like agar.



Definitely not something I always knew and I'd cut through thin stipes going for triangles.

I definitely did cut a fruit from a cluster in half and make plates from the top and bottom of the fruit. I did 3 full-size tubs with the bottom tissue sample and averaged 5.85 oz first flush. Ran a bunch more on the top tissue and got 5.13. Nothing conclusive by any means, just sharing.

Wherever the molds grows is a great indicator of where your problem is.

Ie mold on top grains of jar filter/ lid failure. Or in your case a small hole on the bottom of the bag.

i really dig your approach to logging your clone projects. wish i could be so meticulous.

you know that feeling when you have a mile long list of albums you want to get, then immediately forget it the second you walk into the record store? that's me and mushcult. in the moment, i can't remember a thing i was trying to do.

oh, and thank you rickyswamps for suggesting how to grab clone tissue from the thin stipes. you were totally right.

Yes, stare is right on with smaller samples... I find this to be true for cloning and agar transfers... only takes a microscopic bit of culture to inoculate recieving media, and you have a better chance of a clean break... This is particularly true with clones IME, seems larger tissue samples hold water too long and delay recovery/promote bacteria... just :2cents: though :shrug:

My PC fell over (not while in use) and the pressure gauge broke. I just replaced it but noticed there is a dent where it goes and so it sits looking a tiny bit downward
  It doesnt seem to be significant enough to effect anything but I dont know :undecided: what do you think?

as long as the threads aren't damaged/bent/warped it should still make a tight seal. Was it still very tight fitting when you put it in?

and are you sure it didn't jump a thread and just go in crooked? Maybe back it out and double check it's in line right.

Okay good plan, thank you. I quickly put it on on my lunch break so I will go back later and check again. It is for sure bent but yeah that might not be the reason it's sitting weird. It does fit as snug as the last gauge did
