My best guess is that he knows it's a PITA to drop a single drop onto agar and he knows more than a drop = potential problems. I think bod favors using a sterile shot glass with some spore solution and a loop. IDK but if I had to work with a spore syringe I would probably do the same.

I find without a sharp it is very easy to get one drop on a plate.

Here is what a fraction of a single drop, or barely touching a print is capable of on agar

So if using a syringe how do you go from spore solution to agar? With a loop? Iβm sure you just work from prints at this point just curious. Iβm having shit luck germinating spore syringes to regular plates. I donβt end up with bacteria I just end up with nothing at all lol

Spread plate with inoculation loop is standard procedure for liquids to agar. Some people do a drop right on a plate. I think its horrible practice

I'm mixin' poo and reading old teks. I've always added verm for texture, but older teks just use straight pasteurized poo. Is fluffing it up uneccesary?

i used it for a while ,what i did was i passed it through my straw shredder and it comes out like fine dirt , you can use a blender with water i did that too, then drain and add verm to adjust moisture , its kinda is fluffy after the shred

What's the shot glass for? The spores are the vector, more on the plate = worse time. A loop is flamed

For dipping apparently.
Quote:
bodhisatta said: Have a sterilized shot glass or a blank petri dish(inside is sterile still) flame your needle on the spore syringe then move into the SAB squirt out a drop to cool the needle and then squirt a little bit into your dish take your inoculation loop (I use disposable plastic ones) otherwise flame it bring into the SAB then dip in agar(if flamed) then dip in the spore solution in the dish then make a Z pattern on the dish with the inoculation loop
Sounds good to me.

Say you harvest a shoebox by floating the substrate to the top in water (bods harvest Tek), would the side/bottom pins lose much potency if left for too long harvesting the top since they are water soluble?

Funny i Don't think ive ever used a shot glass to do that. Probably wrote it that way to help noobs along. Squirt right on to the loop
