Do you have the arm holes facing upward with you in a chair and the SAB on the floor or something? I don't get how you can feel your breath on yourself. I always stand for sterile work (going to try and sit from now on, my back is fucked up) and my head is hovering over the top. That + I wear the surgical mask and it's fine; no issues blowing my utensils around.
Put a flame inside the SAB (with zero iso) and move around while breathing normally. Your breath should move the flame around if it's actually hitting you and not the ghost of all the plates you breathed satellites into.
Holding your breath would just cause deeper and more aggressive breaths after. Learn to breathe normally and you'll realize that it ain't that serious. Once you connect your SAB work just a normal routine, no stress, it's easier and easier. That gas mask is overkill, for sure, IMO.

No i have the sab on a counter while I stand. IDK dude, my nose holes point down where my arms are and I can feel it. Maybe you're more over top the sab than I am? I can only get so close to it and can't really lean forward(bent) for too long, but I'm still looking in through the top while working. It's not like my face is in front of the sab holes.

I mean if you're really concerned about your breathing wear a bandana or something to muffle it. But idk I say comfortable is king for sab work

With my little plain surgical mask, there's no disturbance. I feel like I breathe so light anyway that I'd be good. I'm definitely looking into the top as well and not angled much. Can you find another counter that's a little lower?
I mean, do you have a huge contam rate or something? If it ain't broke, don't fix it.

I don't have a lower surface now. When I finish setting up the new room I think the shelf I'd be using is a bit lower.
No I'm not having any issues. This is just kind of a tangent we all got on cause I was curious if anyone else wears a respirator instead of a paper mask and well now here we are.

That's why I kept using it. I never liked the paper masks and bandanas are always too loose or too tight, I can't stop fiddling with them; but my respirator is nice and comfy for me.

I know it's a lot to read through but yes I breathe through my nose. I got out the sab stood like i work and breathing through my nose I could feel the air down my arms, to my hands in the sab.
I appreciate everyone trying to help and make suggestions, but it's not like I'm really having an issue. Was just a curiosity question to start.

Why wear condoms? I don't, and it's fine.

Fun fact, over 50% of the (US) adult population has herpes (HSV-1 or HSV-2).

stareatclouds said: What size of Whatman do you have on there? No other modifications on the lid, such as SHIP or anything? Plastic or metal?
I really think I should write a tek on this because I've dialed it in a lot over the years. I saw the advantages of LC early on and never looked back, and have got it to the point where contamination isn't a concern so much anymore. I've gone through many iterations for LC jar lids. I use a larger size Whatman filter, not sure if it's critical to use a larger filter, but I like to think it helps. They are 50mm 0.3 micron filters. In retrospect it's probably not the filter letting contams in as much as a leak somewhere in the lid. Either way, tinfoil has helped with that. I had a hell of a time drilling holes in the lids and some of the holes got a little bigger than I planned. I use plastic lids from amazon. They seem to seal up pretty good with a strong twist. One trick I finally figured out on the last lid was to use a dremel tool to get the hole to the desired size rather than attempting to drill with a drill bit which resulted in many cracked lids.
Because I am using larger filters I was also able to slip a small piece of silicone tube over the inside vent on the filter and I stuff a small wad of polyfil into that tube. This modification helps prevent slashing LC from entering the whatman filter and soaking the SFD inside.
I use SHIPs, and I don't replace them often at all but I always try to line the syringe up directly with the already existing hole, so that I'm not piercing the same piece of rubber in a dozen places. I don't trust them as a vector so I usually spray them with alcohol right before filling syringes. Keeping tinfoil over the top has been a great help.
For syringes I have a totally different way of reusing them. I have 60ml syringes with 18 gauge needles. I simply place them empty into my pressure cooker and get them to 15psi for 30 minutes. They have been used dozens of times and are all filthy but I don't care.
I use 1 gallon jars. I add 5 grams agar to every 2 liters LC. This gels it up slightly, allowing it to be shaken into a slurry that suspends bubbles. It's possible that these bubbles help provide extra oxygen, although I'd have to do some proper chemistry calculations to determine whether it was a significant. It also changes the way contaminants show up in the LC. I can always spot contams in the LC without needing to test on agar. Bacteria contam will give a yellow hue to the surface, and molds will grow out and even sporulate visibly within the LC. Healthy mycelium will colonize it in a very distinct way and you can tell when it's ready when you see the telltale ice crystal looking growths all throughout the gel stuck to the walls of the jar. Contaminated jars either won't develop those "ice crystals" or will show some obvious sign of contamination.
Unlike typical LC, you don't get isolated clumps of growth filling only 1/3 of the jar of LC. Instead you get completely homogenized distribution of suspended mycelium in each syringe. Colonization occurs uniformly throughout the bag or jar and initiates on every grain all at the same time.
Main reasons why I will never stop using my LC technique: -less work than prepping a hundred plates of agar on a regular basis -I can make enough innoculant that I never need to grain to grain transfer
