Yes. It's the basis behind the entire edible industry. Cubes are no different.
I agree....but I also disagree. Think in nature. Edibles have long lives in nature. Cubes don't. Listen to nature.
In the end it is far worse for the line to keep going back to spores.

Indeed. PE is a great example of a degraded lineage.

So I've inoculated 3 different strains with agar.
I also still have mycelium on agar from those inoculation plates.
Make more pasty plates, keep transferring it, keep it going while I wait on this flush. 5, 10 transfers later to make more grain, keep a chunk of that myc, transfer it, then surely at the point of 10-15-20 transfers, l be near an isolate/monoculture?
Is there a good guess to the number of transfers before reaching the "singular" genetics? What's the proper term on that?
Thanks for all the help everyone.

I have a plate of pe thats 8 months old and i dont have an isolate you can get one in maybe 20 or 10000 just depends on how small of a transfer and how meticulous you are

You won't see sectors for at least 8-10 transfers depending on how many spores you used and how small your transfers are. Could be 20 like cron said.

"Singular genetics" is called different ways, monocultures, isolate, pure culture...

I replied to you in this same thread some days ago, I'm pretty sure you'll get the idea if you read it again.

Josex said: Moab, when you play with agar you can choose lots of ways to do things. This is what I'll do as soon as I have my new apartment set up; I'm going to start with two varieties (mexican and AA+). I'm gonna noc 10 plates from prints for each variety (20 in total). Transfer leading edge from each plate to new plates, and I'm going to be repeating this until I see nice and organized growth (maybe 3 or 4 transfers).
Once I have nice growth in the 20 plates I'm gonna go ahead and and choose a number of them to noc some grains, BUT before that I'll transfer from those plates to new plates to keep isolating further, while at the same time I can have some "limited MS" grows going.
My goal is to keep isolating until I have 20 pure cultures to test out and in the meantime I'll be using donor plates to keep some monotubs going. I'd rather work with isolates than clones, and in the meantime grow MS.

Why start with 20 plates from spores. I would start with 2 or 4 plates from spores than transfer to more plates frome those. No point in making a ton of plates befor you can even see sectoring.

It don't take long to get out of hand. I went through my plates today and threw out close to 100. Moste of them had invitro fruits. About half of them had dropped spores.

Amazon. SEOH is the seller. EU complicates things when it comes to shipping, I'm in the US.

Hopefully it's not as hard for you to get them in Spain as it is in Canada. After shipping I pay like $3 per petri. I just used my last sleeve of 20. I'm sticking with pasty plates from here on out.

Qft... when i first started i had 6 types of cubes with 3 plates each its a hassle it was hard to stay on top of transfers
